摘要
目的探讨脾补肺膏治疗慢性阻塞性肺疾病(COPD)小鼠模型的临床疗效。方法将40只SPF级C57BL/6小鼠随机分为5组,分别为空白组、COPD模型组、健脾补肺膏高剂量组、中剂量组和低剂量组。除空白组外,其余小鼠均采用烟熏结合气管内脂多糖(LPS)滴入法制作COPD小鼠模型,待模型稳定后,空白组和模型组不予以药物治疗,健脾补肺膏高、中、低剂量组小鼠灌服健脾补肺膏溶液(给药浓度分别为20.8、10.4、5.2 ml/kg/d)。观察各小鼠精神状态、饮食情况、体重变化及大小便情况等,检测各组小鼠吸气阻力(RI)、肺静态顺应性、功能残气量(FRC)、趋化因子(KC)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)值。结果空白组和健脾补肺膏高剂量组小鼠体重随饲养周期增大,但健脾补肺膏高剂量组小鼠体重增加较缓慢,而其余3组小鼠体重均有不同程度减轻;COPD模型组小鼠RI值、肺静态顺应性、FRC值较空白组均明显增高,差异均有统计学意义(均P<0.05);健脾补肺膏高、中、低剂量组小鼠RI值、肺静态顺应性以及FRC值均较COPD模型组明显降低,且健脾补肺膏高剂量组小鼠RI值、肺静态顺应性以及FRC值均明显低于健脾补肺膏中、低剂量组,呈剂量依赖性,差异均有统计学意义(均P<0.05);COPD模型组小鼠炎性细胞总数、IL-6、TNF-a、KC值较空白组均明显增高,差异均有统计学意义(均P<0.05);健脾补肺膏高、中、低剂量组小鼠炎性细胞总数、IL-6、TNF-a、KC值均较COPD模型组明显降低,且健脾补肺膏高剂量组小鼠炎性细胞总数、IL-6、TNF-a、KC值均明显低于健脾补肺膏中、低剂量组,呈剂量依赖性,差异均有统计学意义(均P<0.05)。结论健脾补肺膏方能显著增加COPD模型小鼠体重,改善肺功能,同时还能减轻炎性反应。
Objective To investigate the clinical effect of spleen tonifying lung paste in treating chronic obstructive pulmonary disease in mice.Methods 40 SPF C57BL/6 mice were randomly divided into 5 groups,namely control group,COPD model group,jianpibufei ointment in high dose group,middle dose group and low dose group,except the blank group,the remaining mice were exposed to tobacco smoke by intratracheal instillation of lipopolysacchadde (LPS) COPD mice model,to be the model is stable,not to the blank group and the model group medication,jianpibufei cream high in the low dose group of mice orally jianpibufei paste solution(drug concentration were 20.8,10.4,5.2 ml/kg/d).To observe the mice mental state,diet, body weight changes and urine,were detected inspiratory resistance(RI), static pulmonary compliance,functional residual capacity (FRC),chemokine(KC),tumor necrosis factor alpha (TNF- alpha) and interleukin-6 (IL-6) value.Results The blank group and the weight of spleen and lung extract high dose group of mice with feeding period increase,but jianpibufei ointment in high dose group mice increased more slowly, while the remaining 3 groups of mice had different degrees of weight Ioss;COPD model group,RI value,static lung compliance,FRC values were significantly higher than the control group,the difference there was statistically significant (P〈0.05);jianpibufei paste,high medium and low dose group of mice RI,static lung compliance and FRC were significantly higher than COPD model group decreased,and jianpibufei ointment in high dose group of RI value,static lung compliance and FRC value were significantly lower than that of tonifying spleen lung extract medium and low dose group,dose dependently, the differences were statistically significant(P〈0.05);COPD model group,the number of inflammatory celIs,IL-6,TNF-a,KC values were significantly higher than the control group,the differences were statistically significant(P〈0.05);jianpibufei paste,high medium and low dose group of mice the number of inflammatory cells,IL-6, TNF-a,KC values were lower than COPD model group decreased obviously, and jianpibufei ointment in high dose group of total inflammatory cells,IL-6,TNF-a,KC values were significantly lower than that of jianpibufei ointment in the low dose group,dose dependently, the differences were statistically significant(P〈0.05).Conclusion Invigorating spleen and tonifying lung paste can significantly increase the body weight of COPD mice,improve lung function,but also reduce the inflammatory response.
