以AQP表达验证“肺病及肠与泻肠清肺”关链的“水湿”病理证据 被引量:10

Value of AQP Expression to Explain the Significance of Water-dampness Syndrome in the Pathological Mechanism of "Disease of Lung Affecting Intestine/Purging Bowel to Clear Lung"

摘要 目的观察小承气汤对支气管炎模型大鼠肺与结肠组织水通道蛋白(aquaporin,AQP)的调节效应,探讨"肺病及肠与泻肠清肺"之"水湿"关链的可能机制。方法 SD大鼠60只,随机分为空白组、模型组、观察组,每组20只。模型组及观察组单纯烟熏法70天建模。检测大鼠肺功能、血气分析,计算结肠湿重指数,免疫组化法、Western blot法检测大鼠肺组织AQP1蛋白与结肠组织AQP4蛋白表达。结果与空白组比较,模型组大鼠肺顺应性降低、气道阻力增加(P<0.05),肺功能降低,血气分析中PO2降低、PCO2升高(P<0.05),大鼠肺组织AQP1及结肠组织AQP4表达明显下降(P<0.05);与模型组比较,观察组大鼠气道阻力下降,肺顺应性升高(P<0.05);低氧高酸状态(PO2/PCO2/p H)显著改善(P<0.05),肺组织AQP1表达显著升高(P<0.05)。结论肺组织AQP1和结肠组织AQP4可能是"肺病及肠"病理传变中"水湿"的链点之一,亦是"泻肠清肺"功效的新位点,属"肺与大肠相表里"的新证据。 Objective To observe the effect of Xiaochengqi Decoction (XD) on the AQP expression in the lung and the colon of bronchitis rats, thus to reveal possible mechanism of the pathological transfer in the disease of lung affecting intestine as well as purging bowel to clear lung. Methods Totally 60 SD rats were randomly divided into the blank group, the model group, the observation group, 20 in each group. The model of bronchitis was established in the model group and the observation group for 70 days. Pulmonary function and blood gas analysis were detected. The colon wet weight index was calculated, expressions of AQP1 in lung tissue and AQP4 in colon tissue were also determined by immunochemistry and Western blot. Results Compared with the blank group, lung compliance decreased, and respiratory resistance was increased significantly, lung function decreased, PO2 was reduced, PCO2was elevated in blood gas analysis in the model group (P 〈0.05). The expressions of AQP1 in lung tissue and AQP4 in colon tissue obviously decreased (P 〈0.05). Compared with the model group, the respiratory resistance decreased, and the lung compliance were enhanced, the state of hypoxemia and hypercapnia (PO2/PCO2/pH) were also improved (P 〈0.05), and the expression of AQP1 in lung tissue was increased (P 〈0.05) in the observation group. Conclusions AQP1 of lung tissue and AQP4 of colon tissue might be one of connection points involved in the pathophysiology of "disease of lung affecting intestine". They might also be new targets to explain "purging bowel to clear lung". AQP might be new evidence for "interior-exterior correlation between lung and large intestine".
