消银解毒颗粒对Jurkat细胞异常增殖模型的影响 被引量:1

Effect of Xiaoyin Jiedu Granules(消银解毒颗粒) on Model of Jurkat Cell Abnormal Proliferation

摘要 目的探讨消银解毒颗粒治疗银屑病的可能作用机制。方法以植物血凝素(PHA)和白细胞介素6(IL-6)共同诱导Jurkat细胞模拟银屑病T细胞异常增殖的病理模型,制备择取最优效药理血清浓度作用于该细胞模型,采用细胞活性试剂盒(CCK-8)法检测细胞活力,用流式细胞术检测细胞凋亡。结果当药理血清浓度为20%时,消银解毒颗粒组、凉血方组细胞抑制率显著高于解毒方组、阿维A组、蒸馏水组(P<0.01);消银解毒颗粒组细胞早期凋亡率最高,高于凉血方组、解毒方组、阿维A组、蒸馏水组(P<0.05或P<0.01)。结论消银解毒颗粒及其拆方凉血方、解毒方对银屑病T细胞异常增值的炎症病理模型均有抑制作用,且消银解毒颗粒和凉血方的抑制效果明显优于阿维A。 Objective To explore the possible mechanism of Xiaoyin Jiedu Granules (消银解毒颗粒) in the treat- ment of psoriasis. Methods The Jurkat cell was administered by both phytohemagglutinin (PHA) and interleukin 6 (IL-6) to induce the model of psoriasis T cell abnormal proliferation. The optimal pharmaceutical serum concentration was made and selected to be applied to the cell model. The cell viability was measured with cell viability kit (CCK-8), and the apoptosis was detected with flow cytometry. Results When the pharmaceutical serum concentration was 20% , the inhibition rates in the Xiaoyin Jiedu Granules group and Liangxuefang group were significantly higher than that in the Jiedufang group, Acitretin group and distilled water group ( P 〈 0. 01 ) ; the Xiaoyin Jiedu Granules group had the highest early apoptosis rate, which was significantly higher than that in Liangxuefang group, Jiedufang group, Acitretin group and distilled water group ( P 〈 0. 05 or P 〈 0. 01 ). Conclusion Xiaoyin Jiedu Granules and its decomposed recipes could inhibit the inflammatory pathological model of psoriasis T cell abnormal proliferation, and the inhibitory effects of Xiaoyin Jiedu Granules and Liangxuefang seem better than Acitretin.
