摘要
为考察口含型无烟气烟草制品烟碱在口腔黏膜中的渗透行为,设计了透皮扩散池与猪口腔黏膜配套装置,结合高效液相色谱(HPLC)法,研究了口含烟所受压力、口含烟pH及含水率对其所含烟碱渗透速率的影响。结果表明:(1)在0.70~44.65μg/m L质量浓度范围内烟碱标准曲线的线性良好(R^2=1.000),回收率为96.45%~98.96%;(2)口含烟中烟碱透过猪口腔黏膜为直线规律,属于膜-贮库型;(3)在0.5~12.0 h内,施加6.65×10^(-2)N/cm^2的压力可显著提高烟碱的渗透速率;(4)含水率处于30%~50%之间时,pH=7.4~8.4的口含烟的烟碱渗透速率明显高于pH=5.3~5.4的口含烟。因此,可主要通过改变压力和口含烟pH来调控口含烟烟碱在口腔黏膜中的渗透行为。
In order to investigate the nicotine permeation behavior in oral mucosa from snus, a device was designed which incorporated a matching skin permeation diffusion cell and a pig's oral mucosa. The effects of pressure, pH and moisture content on the nicotine permeation rate from snus were studied by combining the designed device with high performance liquid chromatography. The results showed that: 1) R2 of the standard curve of nicotine concentration was 1.000 in the range of 0.70-44.65 μg/mL and the recovery ranged from 96.45% to 98.96%. 2) Nicotine permeation from snus through the pig's oral mucosa as time presented a linear relationship, which belonged to a mucosa film-storage type. 3) Within 0.5-12.0 h, applying a pressure of 6.65× 10^-2 N/cm^2 significantly increased the permeation rate of nicotine. 4) When the moisture content of the snus sample was between 30% and 50%, the permeation rate of nicotine from snus at pH of 7.4-8.4 was significantly higher than that at pH of 5.3-5.4. Therefore, the nicotine permeation behavior of snus in oral mucosa could be regulated by changing the pressure and pH of snus.
