白藜芦醇在复杂性区域疼痛综合征Ⅰ型大鼠模型中的作用及机制 被引量:1

摘要 目的:探讨白藜芦醇对复杂性区域疼痛综合征Ι型(CRPS1)大鼠模型的疗效及作用机制。方法:成年雄性SD大鼠50只分为正常对照组(A组)、假手术组(B组)、模型组1(C组)、模型组2(D组),模型组3(E组)。A、B、C 3组予5%DMSO溶液,D组予MIF抑制剂ISO-1 1 mg/(kg·d),E组予白藜芦醇20 mg/(kg·d);连续腹腔注射14 d。第0、7、14天测疼痛行为,血MIF、TNF-α浓度;第14天测坐骨神经中MIF、ERK1/2和p-ERK1/2蛋白的表达。结果:第7及14天,D、E组疼痛阈值较前明显改善,血清MIF、TNF-α水平下降,坐骨神经MIF及p-ERK1/2蛋白表达明显下调,且显著低于C组(P<0.05),第7天血清MIF、TNF-α已降至正常,第14天大鼠疼痛阈值也降至正常,与A组比较无明显差异(P>0.05)。结论:白藜芦醇可显著改善CRPS1大鼠模型的疼痛行为,下调MIF和ERK1/2磷酸化水平的表达,通过抑制MIF/ERK信号通路干预CRPS1大鼠的疼痛行为。 Objective To investigate the role and mechanism of resveratrol in a rat model of complex regional pain syndrome type I (CRPS1). Methods 50 male SD rats were divided into control group (group A), sham operation group (group B) , model group 1 (group C), model group 2 (group D) , and model group 3 (group E). Groups A, B and C received 5% DMSO; group D received ISO-1 of 1 mg/(kg-d) ; and group E received resveratrol of 20 mg/(kg, d) for 14 days. Pain behaviors were assessed on days 0, 7, and 14. Serum levels of MIF and TNF-α were detected by ELISA. MIF, total ERK1/2 and p-ERK1/2 in sciatic nerve were detected by Western blot. Re- sults On days 7 and 14 after treatment, resveratrol injection, similar to ISO-1, significantly improved the pain threshold; serum levels of MIF and TNF-α were significantly decreased; expressions of MIF, total ERK1/2 and p-ERK1/2 in sciatic nerve were also decreased significantly in group E, which were significantly lower than group C (P 〈 0.05). Conclusions Resveratrol can significantly improve pain threshold, decrease expressions of MIF and p-ERK1/2 in a rat model of CRPS1, which might be involved in the inhibition of ERK signaling pathway.
