Determination of Formaldehyde Content in Black Fungus by HPLC

摘要 建立利用高效液相色谱测定黑木耳中甲醛的一种方法。用衍生液提取样品中的甲醛,反应生成甲醛衍生物,液相色谱法测定。色谱条件为:色谱柱C_(18)(150 mm×4.6 mm,5μm)、流动相为乙腈-乙酸钠缓冲液(V/V,50︰50)、流速1.0 m L/min、柱温40℃、检测波长365 nm、进样量20μL。甲醛的线性范围为5~100 ng/m L,线性回归方程为Y=2.915 6X-16.346 4,在鲜木耳和干木耳中加入约为本底值的0.5,1和2倍的甲醛,其平均回收率为87.60%~93.25%。方法的检出限(S/N=3)为2 mg/kg。该方法操作简单,稳定性和重现性好,结果准确度高,适用于黑木耳中甲醛含量的分析测定。 A method was established using HPLC for determination of formaldehyde in the black fungus. The derived fluid was used to extract formaldehyde in the sample, which produced formaldehyde derivatives through the reaction. The derivatives were measured by HPLC. HPLC conditions were column Cls (150 mm × 4.6 mm, 5 μm), mobile phase was acetonitrile- sodium acetate buffer (V/V, 50 : 50); Flow rate was 1.0 mL/min, Column temperature was 40℃, Detection wavelength was 365 nm and injection volume was 20 μL. The linear range of formaldehyde was 5-100 ng/mL, the linear regression equation was Y=2.915 6×-16.346 4, and the formaldehyde add level was about fresh black fungus and dried black fungus background value of 0.5, 1 and 2 times, the average recovery was 87.60%--93.25%. The method limit (S/N=3) was 2 mg/kg. The method is simple, stable and reproducible, with accurate results, and hence it is suitable for determination of Formaldehyde content in the black fungus.
