针刺对慢性疲劳综合征大鼠血浆T细胞转录因子/GATA结合蛋白3表达的影响 被引量:16

Effect of Acupuncture on the Expression of Transcription Factor T-bet/GATA-3 in Plasma of Rats with Chronic Fatigue Syndrome

摘要 目的:观察针刺对慢性疲劳综合征(CFS)大鼠血浆T细胞转录因子(T-bet)/GATA结合蛋白3(GATA-3)表达的影响,探索针刺治疗CFS的作用机制。方法:SD大鼠随机分为空白对照组、模型组、针刺组、人参皂甙组,每组12只。采用束缚+冷水游泳的复合应激法制备CFS大鼠模型。针刺组针刺"百会""关元""足三里",人参皂甙组予人参皂甙水溶液灌胃,均1次/d,14d后,观察各组行为学变化,采用RTPCR法检测血浆T-bet/GATA-3基因的表达水平。结果:模型组大鼠悬挂不动时间长于空白对照组(P<0.05),力竭游泳时间短于空白对照组(P<0.05);与模型组比较,人参皂甙灌胃及针刺治疗后大鼠悬挂不动时间均缩短(P<0.05),力竭游泳时间均延长(P<0.05);且针刺组较人参皂甙组悬挂不动时间更短,力竭游泳时间更长(均P<0.05)。模型组大鼠血浆T-bet基因的表达水平高于空白对照组(P<0.05),GATA-3基因表达水平低于空白对照组(P<0.05);人参皂甙灌胃及针刺治疗均能降低T-bet基因的表达水平(P<0.05),升高GATA-3基因的表达水平(P<0.05);针刺组较人参皂甙组T-bet表达水平降低更显著(P<0.05)。模型组T-bet/GATA-3比值高于空白对照组(P<0.05);人参皂甙灌胃及针刺治疗后T-bet/GATA-3比值较模型组均下降(P<0.05)。结论:针刺能降低血浆T-bet表达水平,升高GATA-3表达水平,调节升高的T-bet/GATA-3比值,可能是其治疗CFS的机制之一。 Objective To observe the effect of acupuncture on the expression of T-box expressed in T ceil (T-bet)/ GATA binding factor-3 (GATA-3) in plasma of rats with chronic fatigue syndrome (CFS) and explore the mechanism of acupuncture treatment for CFS. Methods Forty-eight healthy male SD rats were randomly divided into blank control group, CFS model group, acupuncture group, and ginsenoside group (12 rats in each group). OFS rat model was established by combining restriction and cold water swimming. Acupuncture was applied to "Baihui"(GV 20), "Guanyuan" (CV 4) and "Zusanli" (ST 36, bilateral) acupoints, once a day for two weeks. The ginsenoside group was gavage administrated with ginsenoside, once a day for two weeks. After 14 days, behavioural changes were observed, and the expression levels of T-bet/GATA-3 genes in plasma were detected by RT-PCR. Results Compared with the blank control group, the time for immobility of forced suspensory test was significantly longer (P〈0.05) and the time for exhaustive swimming was significantly shortened (P〈0.05) in the CFS model group. Compared with the model group, the two indexes above-mentioned were reversed (P〈0.05) both in the acupuncture group and the ginsenoside group, and the effects in the acupuncture group were more significant than those in the ginsenoside group (P〈0.05). Compared with the blank control group, the expression level of T-cell transcription factor T-bet gene in plasma was higher in the CFS model group ( P〈0.05), companied with lower GATA-3 gene expression ( P〈0.05). The ratio of T-bet/GATA-3 was higher in the model group than in the blank control group(P〈0.05). Compared with the CFS model group, all the indexes above-mentioned were reversed (P〈0.05) in the two treatment groups. Acupuncture group showed a better effect on reducing T-bet gene expression than the ginsenoside group (P〈0.05). Conclusion Acupuncture can decrease the expression level of T-bet gene while increase the expression of GATA-3 gene, which may be associated with its role in treating CFS.
