血清素受体5-HT3与5-HT4在心肌衰竭鼠中的作用

Effect of 5-HT3 and 5-HT4 on heart failure rat

目的 研究5-羟色胺3（5-HT3）与5-羟色胺4（5-HT4）在心肌衰竭鼠模型中的作用.方法 对Wistar鼠进行左前降支结扎法手术建立心力衰竭鼠模型.用MRI和ECG方法测定左心室功能,并用real-time PCR法检测相关基因的表达并进行相关性分析.结果 在心肌衰竭鼠中,心脏质量、心脏与体质量比值、左心室舒张末期压（LVEDP）和左心房直径（LAD）值均增加;脑钠肽（BNP）和5-HT4 mRNA表达及肌球蛋白重链β（βMHC-β）/肌球蛋白重链α（MHC-α）值上升;但左心室收缩末期压（LVESP）和左室短轴缩短率（LVFS）值下降,5-HT3 mRNA表达减少,差异有统计学意义（P均〈0.05）.5-HT3与MHC-β/MHC呈负相关（r=-0.9173,P〈0.05）,5-HT4与MHC-β/MHC-α呈正相关（r=0.9495,P〈0.01）,5-HT3与5-HT4呈负相关（r=-0.8772,P〈0.05）.结论 在心肌衰竭鼠中,5-HT3 mRNA表达下调和5-HT4上调影响鼠左心室功能,具有成为治疗靶点的潜在意义.

Objective To study the effect of 5-HT3 and 5-HT4 on heart failure rat.Methods LAD ligation method was used to create heart failure rat model.MRI and EGC were used to test left ventricular function.mRNA expressions of relative genes were performed by real-time PCR,and correlation analysis was conducted.Results Compared with sham rats,in heart failure rats,the heart weight,heart weight/body weight,LVEDP and LAD were higher;mRNA expression of BNP and 5-HT4,and MHC-β/MHC-α were increased;however,LVESP,LVFS and 5-HT3 mRNA expression were decreased（P all〈0.05）.MHC-β/MHC-α had a negative correlation with 5-HT3（r=-0.9173,P〈0.05）,while it was positively correlated with 5-HT4（r=0.9495,P〈0.01）.There was a negative correlation of 5-HT3 with 5-HT4（r=-0.8772,P〈0.05）.Conclusions Downregulated 5-HT3 and upregulated 5-HT4 can affect left ventricular function in heart failure rats,and they may have a potential meaning of being therapeutic target.
