利用甲状腺细针吸取细胞学染色涂片检测BRAF(V600E)突变

摘要 目的验证采用细针吸取细胞学(FNA)染色涂片检测BRAF(V600E)突变的可能性。方法对甲状腺患者的FNA染色涂片和手术切除标本的石蜡切片取样进行BRAF(V600E)突变检测进行比较,观察细胞涂片取样方法的敏感度、特异性、准确率以及两种取样方法所得DNA的质量差异。结果共41例患者同时检测了FNA染色涂片和石蜡切片的BRAF(V600E)突变。敏感性为96.9%(31/32),特异性为85.7%(6/7),准确率为90.2%(37/41)。将阳性病例的ΔCt值进行配对的t检验,t=5.567,P<0.0001,表明染色涂片的DNA质量要优于石蜡切片。结论利用甲状腺FNA染色涂片能有效检测到BRAF(V600E)突变检测。 Objective Detection of BRAF (V600E)mutation through thyroid fine needle aspiration (FNA)stained smears is validated in this study. Method BRAF(V6OOE) mutation tests are carried through thyroid FNA fresh direct stained smears and paraffin sections of subsequent resection specimen to observe the sensitivity, specificity, accuracy of new sample method and the DNA quality difference between two methods. Result BRAF(V60OE) mutations are concurrently detected by stained smears and paraffin sections for 41 cases. The sensitivity is 96.9% (31/32) , the specificity 85.7% (6/7)and the accuracy 90.2% (37/41). ACt values of BRAF (V600E)mutation test from the positive cases is checked by Paired Sample t Test, t = 5. 567,P〈0. 0001, which demonstrates that the DNA quality from stained smears is better than from paraffin sections. Conclusion BRAF (V60OE)mutation can be efficiently de- tected by FNA stained smears.
