滋阴补肾方对围绝经期综合征患者外周血单个核细胞ERK及kisspeptin分子的影响

Effects of Ziyin Bushen Recipe on ERK and Kisspeptin Molecules in Peripheral Blood Mononuclear Cells of Patients with Perimenapausal Syndrome

目的:观察滋阴补肾方对围绝经期综合征(PMS)患者外周血单个核细胞细胞外调节蛋白激酶(ERK)及kisspeptin分子的影响,以期探讨该方可能的分子作用机制。方法:选择符合评价条件的肾阴虚型PMS患者104例,随机分为治疗组和对照组;治疗组服用滋阴补肾方,对照组服用更年安片,疗程均为8周。治疗结束,评价两组的综合临床疗效,并检测患者治疗前后外周血单个核细胞ERK、磷酸化ERK(pERK)及kisspeptin等指标变化。结果:两组在综合疗效方面无显著性差异(P>0.05),但治疗组在证候改善方面优于对照组。治疗组可明显下调外周血pERK和kisspeptin-10水平(P<0.01),而对ERK无明显影响;对照组可下调pERK水平(P<0.05),但对ERK和kisspeptin-10均无明显影响(P>0.05)。治疗组对于pERK和kisspeptin-10的调节作用优于对照组(P<0.01或P<0.05)。结论:滋阴补肾方可能通过上游途径,以kisspeptin-10和pERK分子为介导而发挥对下丘脑-垂体-卵巢轴的调控作用。

Objective ： To observe the effects of Ziyin Bushen Recipe on extracellular regulated protein kinase （ERK） and kisspeptin molecules in peripheral blood mononuelear cells of patients with perimenapausal syndrome （ PMS）, and discuss its possible molecular mechanism. Methods： 104 PMS patients with syndrome of kidney yin deficiency who met the evaluation criteria were selected and randomly divided into the treatment group and control group. The treatment group was treated with Ziyin Bushen Recipe, and the control group was treated with Gengnian＇ an tablets, with a course of 8 weeks. After treatment, the comprehensive clinical efficacy in the two groups was evaluated, and the changes of ERK, phosphorylated ERK （pERK）and kisspeptin molecules in peripheral blood mononuclear cells were detected. Results： There was no significant difference on the comprehensive efficacy between the two groups （ P 〉 0.05 ）, but the improvement on syndrome in the treatment group was better than that in the control group. The levels of pERK and kisspeptin-10 in peripheral blood were significantly down-regulated in the treatment group（ P 〈 0. 01 ）, but no obvious change of ERK was found. The pERK level was down-regulated in the control group（P 〈 0.05 ）, but there were no significant effects on ERK and kisspeptin-10 （P 〉 0. 05 ）. The effects on pERK and kisspeptin-10 in the treatment group were better than those in the control group （P 〈 0.01 or P 〈 0.05 ）. Conclusion： Ziyin Bushen Recipe might play a role in regulating hypothalamic-pituitary-ovarian axis function through upstream pathway mediated by kisspeptin-10 and pERK.
