摘要
目的:研究鹿红颗粒对心力衰竭大鼠心脏指数(HI)、左室质量指数(LVMI)、脑钠肽(BNP)及心肌结构的影响,以探讨其对心力衰竭大鼠心功能的保护作用。方法:采用腹主动脉缩窄术制备高血压后心力衰竭大鼠模型。大鼠随机分为假手术组、模型组、雅施达组及鹿红颗粒低、中、高剂量组,除假手术组外均造模。相应药物灌胃干预8周后计算大鼠HI、LVMI;采用ELISA方法测定大鼠BNP水平;采用HE染色、透射电镜下分别观察大鼠心肌细胞病理形态结构及超微结构改变。结果:与模型组比较,鹿红颗粒高、中、低剂量组均能显著降低LVMI、HI(P<0.01);鹿红颗粒高、中、低剂量组与模型组相比,均能有效降低大鼠BNP(P<0.01),鹿红颗粒高剂量组优于中剂量组(P<0.05)及低剂量组(P<0.01)。通过对大鼠心肌HE染色、透射电镜观察发现,与模型组比较,鹿红颗粒各组均能明显改善心肌细胞病理形态结构及超微结构。结论:鹿红颗粒能改善心力衰竭大鼠心室重构、减轻心肌损伤、提高心功能。
Objective: To study the influence of Luhong Granule on heart index ( HI ), left ventricular mass index (LVMI) ,brain natriuretic peptide(BNP) and myocardial structure in heart failure rats,and discuss its protective effects on heart function of rats with heart failure. Methods: Rat model of hypertensive heart failure was established by abdominal aortic coarctatiou. After successful modeling, the rats were randomly divided into the sham operation group, model group, perindopril group, Luhong Granule groups with low-, middle-, high-dose. After the corresponding intervention for 8 weeks, the HI and LVMI of rats were calculated, the BNP level in rats was detected by enzyme-linked immunosorbent assay, the morphological structure and uhrastructure of myocardial cells were observed by HE staining and transmission electron microscopy respectively. Results: Compared with the model group, the LVMI, HI and the BNP level were significantly decreased in Luhong Granule groups with low-, middle-, high-dose ( P 〈 0.01 ). The effects of the high-dose group were better than those of the middle-dose group ( P 〈 0.05 )and low-dose group( P 〈 0.01 ). By HE staining and transmission electron microscopy, the morphological structure and ultrastructure of myocardial cells were obviously improved in Luhong Granule groups with low-, middle-, high-dose compared with the model group. Conclusion: Luhong Granule can improve the ventricular remodeling in heart failure rats, reduce the myocardial damage and improve the heart function.
