针刺对慢性高眼压后兔模型视网膜细胞凋亡及ERK1和MAPK9的影响 被引量:6

Effects of Acupuncture on Retinal Cell Apoptosis and Expressions of ERK1 and MAPK9 in Rabbits with Chronic Intraocular Hypertension

摘要 目的:探讨针刺治疗对慢性高眼压后兔模型视网膜神经节细胞的保护作用及可能的作用机制。方法:选择新西兰兔,采用随机数字表法设为正常组、模型组、针刺组,其中模型组与针刺组以复方卡波姆诱导建立慢性高眼压后兔模型。造模2周后,针刺组给予毫针合谷(LI 4)、睛明(BL 1)、球后(Ex-HN 07)穴治疗,隔天1次,每周3次,共治疗4周;正常组和模型组不予处理。干预结束后,取各组兔眼球内视网膜组织,采用流式细胞仪测视网膜细胞凋亡情况,并以免疫组化法检测神经生长因子(NGF)信号通路下游通路的细胞外调节蛋白激酶-1(ERK1)、丝裂原活化蛋白激酶-9(MAPK9)蛋白表达。结果:与正常组比较,模型组左上象限晚期凋亡细胞占总细胞的百分比明显增高(P<0.05);与模型组比较,针刺组晚期凋亡细胞占总细胞的百分比明显降低(P<0.05)。与正常组比较,模型组与针刺组ERK1积分光密度水平均明显下降(P<0.01);模型组MAPK9积分光密度水平有所上升,而针刺组明显下降(P<0.05)。结论:针刺作用于慢性高眼压后兔模型,可在一定程度上降低视网膜细胞的凋亡率,且对MAPK下游信号分子MAPK9的表达有减弱作用。 Objective : To discuss the protective effects and possible mechanism of acupuncture on retinal ganglion cells of rabbit with chronic intraocular hypertension. Methods: The New Zealand rabbits were selected and divided into the normal group, model group and acupuncture group according to random number table. Rabbit model of chronic intraocular hypertension induced by compound carbomer was established in the model and acupuncture group. 2 weeks after modeling,the acupuncture group was treated with acupuncture of filiform needle at Hegu ( LI4 ), Jingming ( BL1 ) and Qiuhou ( Ex-HN07 ) acupoints every other day. The treatment course was 3 times a week and totally 4 weeks. The normal and model group were not received any treatment. After intervention, the intraocular retinal tissue of rabbits in each group was collected. The apoptosis of retinal cells was measured by flow cytometry, and the protein expressions of extracellular regulated protein kinase- 1 ( ERKI ) and mitogen activated protein kinase-9 (MAPK9) in downstream nerve growth factor (NGF) signaling pathway were detected by immunohistochemistry. Results: Compared with the normal group, the percentage of late apoptotic cells in total ceils in left upper quadrant of the model group was significantly increased ( P 〈 0.05 ). Compared with the model group, the percentage of late apoptotic ceils in total ceils of the acupuncture group was significantly decreased ( P 〈 0.05 ). Compared with the normal group, the integral optical density level of ERK1 was significantly decreased in the model and acupuncture group( P 〈 0. 01 ). The integral optical density level of MAPK9 was increased in the model group and significantly decreased in the acupuncture group( P〈 0.05 ). Conclusion: Acupuncture effects on chronic intraocular hypertension in rabbits, which can decrease the apoptosis rate of retinal cells to some extent, and decrease the expression of MAPK9.
