参附注射液对大鼠急性肺损伤保护作用的实验研究

Protective Effects of Shenfu Injection on Acute Lung Injury in Rats

目的观察参附注射液对脂多糖性急性肺损伤(ALI)大鼠肺组织的保护作用及探讨其可能的作用机制。方法 54只健康雄性Wistar大鼠按随机数字表法分为对照组(NS组)、急性肺损伤组(ALI组)、参附注射液干预组(SF组),急性肺损伤模型通过股静脉注射脂多糖(5 mg/kg)建立。SF组于造模前30 min给予参附注射液(10 mL/kg),NS组和ALI组均给予等量0.9%氯化钠注射液。分别于用药3 h、6 h、12 h后,观察大鼠肺组织的病理改变,检测血气分析,记录肺组织湿/干重比(W/D),测定大鼠肺组织中肿瘤坏死因子F-α(TNF-α)、白介素-6(IL-6)、白介素-10(IL-10)的表达。结果与ALI组相比,SF组ALI的程度明显减轻,Pa O2显著改善(P<0.05),肺W/D比值降低(P<0.05),肺组织TNF-α、IL-6浓度显著下降,IL-10浓度明显升高(P<0.05)。结论参附注射液对ALI大鼠肺组织具有一定的保护作用,其机制可能是降低了TNF-α、IL-6的浓度,促进IL-10的表达,调节促炎-抗炎平衡,减轻肺部及全身的炎症反应。

Objective： To study Protective effect of Shenfu Injection on lung tissue in rats with lipopolysaccha-ride induced acute lung injury and its possible mechanism. Methods： 54 male Wister rats were randomly divided into the control group（NS group）,acute lung injury group（ALI group）,and Shenfu injection group（SF group）. A-cute lung injury model was established by injection of lipopolysaccharide （5 mg/kg）. 30 min before modeling,SF group was given Shenfu injection（10 mL/kg）,and NS group and ALI group were given equivalent saline. In 3 h,6 h and 12 h,the pathological changes of lung tissue in rats were observed,and blood gas analysis,Wet/dry weight ratio of lung tissue（W/D）,and the expression of TNF-α,IL-6 and IL-10 in rat lung tissue were determined. Re-sults： Compared with ALI group,the degree of acute lung injury was significantly reduced in SF group;PaO2 was significantly improved （P〈0.05）,and the pulmonary W/D ratio was decreased （P〈0.05）;TNF-αand IL-6 con-centrations in lung tissue were significantly decreased and IL-10 concentration was significantly increased （P〈0.05）. Conclusion：Shenfu injection has protective effects on lung tissue of rats with ALI,whose mechanisms may involve decreasing the expression of TNF-α and IL-6,increasing the expression of IL-10,regulating the balance of pro-inflammatory and anti-inflammatory,and reduce inflammation in the lungs and the whole body.
