摘要
目的:探讨丹皮酚、三七总皂苷联合应用对急性心肌梗死(AMI)后心室重构大鼠心肌组织NF-κB p65、TNF-α表达的影响。方法:结扎大鼠左冠状动脉前降支制备AMI动物模型,丹皮酚组、三七总皂苷组、联合用药(丹皮酚+三七总皂苷)高、低剂量组、卡托普利组大鼠分别给予相应药物腹腔注射28d。检测各组大鼠的左心室收缩压(LVSP)、左心室舒张末压(LVEDP)及左心室内压力最大上升和下降速率(±dp/dt_(max)),Western blot法检测心肌组织NF-κB p65、TNF-α蛋白的表达情况。结果:与模型组比较,用药组各指标均明显改善;并且,联合用药组各指标的改善情况明显优于单味药组,联合高剂量组明显优于联合低剂量组。结论:丹皮酚、三七总皂苷联合应用能明显改善心梗后大鼠的心室重构,其机制可能与抑制NF-κB p65、TNF-α蛋白表达有关,且联合用药优于单独用药。
Objective:To investigate the effects of paeonol (Pae) combined Panax Notoginseng Saponins (PNS) on NF-κB p65、TNF-α expression in cardiac muscle of ventricular remodeling rats after acute myocardial infarction (AMI). Methods: The AMI rats' model was made by ligating the anterior descending branch of left coronary artery; And then the rats in Pae group, PNS group, Pae+PNS combination low dose group, Pae+PNS combination high dose group and captopril group were intraperitoneally injected with corresponding drug for 28 days. The left ventricular systolic pressure, left ventricular end diastolic pressure, and maximum rise and fall rate of left ventricular pressure of rats in each group were detected; And Western blot was used to detect the NF-κB p65、TNF-α expression in cardiac muscle of rats. Results: Compared with rats in model group, the indexes of rats in each drug group improved obviously. Moreover, the improvement of indexes of rats in combination group were significantly better than single medicine group; especially the combination high dose group. Conclusions: Pae combined with PNS can obviously improve ventricular remodeling of rats after myocardial infarction, the mechanisms maybe relate to inhibit NF-κB p65、TNF-α expression in cardiac muscle; Moreover, the effects of drug combination is better than single medicine.
