摘要
目的检测SLARP蛋白免疫原性及能否成为疟疾疫苗潜在候选抗原。方法构建pc DNA3.1(+)/Py SLARP真核表达载体并表达,免疫小鼠并检测SLARP蛋白抗体及相关细胞因子水平。结果经pcDNA3.1(+)/PySLARP免疫后的小鼠血清中总IgG、IgG1、IgG2a抗体水平高于对照组,差异有统计学意义(P<0.01),并且小鼠脾细胞悬液细胞因子IFN-γ、IL-2、IL-4和IL-10水平也明显高于对照组,差异有统计学意义(P<0.01)。结论 PySLARP蛋白可以同时诱导体液和细胞免疫应答,具有一定的免疫原性,可以作为疟疾疫苗成分的一种候选抗原。
Objective To test the immunogenieity of the sporozoite and liver stage asparagine-rich protein from Plasmodium yoelii (PySLARP) and the potential candidate antigen for malaria vaccine. Methods Constructed the eukaryotic expression vector pcDNA3.1 (+)/PySLARP. Then, the vector was immunized to BALB/c mice and some indexes about immunogenicity were detected. Results The levels of total IgG, IgG subtypes (IgG1 and IgG2a) and cytokines (IFN-y, IL-2, IL-4 and IL-10) in the experimental group were significantly higher than those in the control groups.Conclusion PySLARP could induce obvious celluar and humoral immune response ; it may be a candidate for plasmodium vaccine.
