摘要
以粤糖93-159甘蔗为试材,对其进行预处理,茎尖初始培养、增殖培养、生根培养,组培苗的炼苗、移栽等研究。结果表明:经预处理,茎段腋芽萌发率达90%;茎尖在MS+6-BA1.0 mg/L+2,4-D1.0 mg/L+AC6.0 g/L培养基中生长良好;在增殖培养基MS+6-BA 1.0 mg/L+KT 0.5 mg/L中增殖系数达9.4;在生根培养基1/2MS+NAA 1.0 mg/L中,生根率达93%;经炼苗移栽存活率为80%。
Using Yuetang 93-159 sugarcane as material, initial culturing, multiplication culturing, root culturing for stem tip, acclimatizing and transplanting for tissue culture seedling were conducted. The result showed that the lateral bud germination rate was 90% after pretreating, stem tip growed well in the culture medium of MS ＋6- BA1.0mg/L＋2,g-D1.0mg/L＋AC6.0g/L, multiplication rate was 9.4 in the subculture medium of MS＋6-BA1.0mg/L＋ KT0.5 mg/L, rooting rate is high to 93% in the rooting medium of 1/2MS＋NAA1.0mg/L, and survival rate was 80% after acclimatizing and transplanting.
