摘要
以血小板衍生生长因子(PDGF-BB)为研究对象,利用核酸适体与其特异性识别作用,采用辣根过氧化物酶(HRP)作为标记物,催化luminol-H_2O_2化学发光体系,通过化学发光成像技术建立PDGF-BB双适体识别生物传感器,实现对PDGF-BB的高灵敏检测。线性范围是0~10nmol·L^(-1),检测限0.87nmol·L^(-1);且具有良好的选择性,可实现人体血浆等复杂样品中PDGF-BB的特异性检测。
This study developed a chemiluminescence imaging assay (CLI) of platelet-derived growth factor-BB (PDGF-BB) based on its specific recognition to dual-aptamer.In this system, horseradish peroxidase (HRP) was employed as labels.The resulting CLI biosensor achieved sensitive detection of PDGF-BB with a linear range of 0 to 10 nmol·L^-1 and a detection limit of 0.87 nmol·L^-1.In addition, this CLI biosensor exhibited good selectivity, achieving specific detection of PDGF-BB in complex sample, such as human serum.
