摘要
目的:研究白芥子涂法穴位贴敷对哮喘豚鼠的抗气道高反应性作用,并初步探讨其机制。方法:将豚鼠分为正常组、哮喘组、贴敷治疗组。模型采用OVA致敏法,首次激发后,行每周一次的白芥子涂法贴敷,共3次。末次激发后24 h,小动物呼吸功能测试仪检测呼气阻力、吸气阻力、肺顺应性,HE染色检测肺组织炎症,免疫组化法检测肺组织TRPV1的定位与表达,免疫印迹法检测肺组织TRPV1蛋白的表达水平。结果:与正常组相比,哮喘组豚鼠呼气阻力、吸气阻力明显上升,肺顺应性明显下降,TRPV1表达略有上升(P<0.05);与哮喘组相比,贴敷治疗组豚鼠哮喘组豚鼠呼气阻力、吸气阻力显著下降,肺顺应性上升,TRPV1表达显著下调(P<0.05)。结论:白芥子涂法穴位贴敷可能通过调节肺组织TRPV1,降低哮喘中的气道高反应性感性。
Objective : To study the anti - airway hypersensitivity of the Baijiezi Tufa to asthmatic guinea pigs and explore its mechanism. Methods:The guinea pigs were divided into normal group, asthma group, acupoint application treatment group and OVA sensitized bronchial asthma model. After the first challenge,the guinea pigs were treated with Baijiezi Tufa three times per week. At the end of 24 hours after the last challenge, the respiration rate, inspiratory resistance and lung compliance were meas- ured by small animal respiration tester. The lung tissue inflammation was detected by HE staining. The localization and expression of TRPV1 in lung tissue were detected by immunohistochemistry and Western Blot in lung. Results : Compared with the normal group, the bronchial resistance and inspiratory resistance of asthmatic guinea pigs increased significantly. The lung compliance de- creased and the expression of TRPV1 increased slightly. Compared with the asthma group,inspiratory resistance decreased signifi- cantly( P 〈 0. 05 ), lung compliance increased and TRPV1 expression was significantly reduced( P 〈 0. 05 ). Conclusion:Acupoint application of Baijiezi may reduce the airway hyperresponsiveness of asthma by regulating TRPV1 in lung tissue.
