摘要
目的:探讨清化颗粒对db/db小鼠肠道胰高血糖素样肽-1(GLP-1)合成和分泌的影响。方法:选取db/m小鼠为空白对照组,db/db糖尿病小鼠随机分为模型对照组和清化颗粒(低、中、高剂量)组,干预4周,观察小鼠糖代谢变化;ELISA法分析小鼠血清GLP-1的水平;qRT-PCR法检测小鼠回肠胰高血糖素原和激素原转化酶1/3(PC1/3)的mRNA水平;Western blot法检测小鼠回肠PC1/3的蛋白表达水平;组织病理学观察小鼠回肠组织形态变化。结果:与模型对照组比较,清化颗粒降低db/db糖尿病小鼠的空腹血糖(FBG)(P<0.01);提高血清GLP-1的浓度(P<0.01);上调小鼠回肠组织胰高血糖素原和PC1/3酶的mRNA水平(P<0.01);上调小鼠回肠组织PC1/3的蛋白表达水平(P<0.01);并具有剂量依赖效应。另外,清化颗粒明显改善db/db糖尿病小鼠回肠组织形态。结论:清化颗粒能够降低db/db糖尿病小鼠的血糖水平,提高血清GLP-1水平,上调胰高血糖素原和PC1/3的mRNA水平,促进肠道GLP-1的合成和分泌,改善回肠组织形态。
Objective : To investigate whether Qinghua granule (QHG) affects GLP - 1 release in db/db mice. Methods : Db/m mice were as normal control group. Db/db mice were randomized into model control group, QHG ( low, medium, high - dose) groups. All the groups were administrated for 4 weeks. Blood glucose was detected. The serum concentration of GLP - 1 was tested by ELISA. The mRNA levels of proglucagon and prohormone convertase 1/3 (PC1/3) in ileum of db/db mice were detected by qRT - PCR. The protein level of PC1/3 in ileum of db/db mice were detected by Western blot. The structure of ileum tissue was observed by histopathological analysis. Results : Compared with model control group, QHG exhibited dose - dependent effect. QHG could decrease FBG and increase the concentration of GLP - 1 in serum ( P 〈 0. 01 ). QHG up - regulated the mRNA level of pro- glucagon and PC1/3 in ileum tissue significantly (P 〈 0. 01 ). QHG up - regulated the protein level of PC1/3 in ileum tissue sig- nificantly (P 〈 0. 01 ). In addition, QHG obviously improved the structure of ileum of db/db mice. Conclusion:QHG could reduce FBG obviously, increase GLP - 1 concentration in serum, up - regulate the mRNA level of proglucagon and PC1/3, affect GLP - 1 release and improve the structure of ileum in db/db mice.
