薯蓣丸对豚鼠变应性鼻炎模型Foxp3的影响

Effect of Dioscorea opposita Thunb. pills on Foxp3 in guinea pigs with allergic rhinitis

目的:观察薯蓣丸对豚鼠变应性鼻炎模型Foxp3的影响。方法:将60只豚鼠随机分为空白组,薯蓣丸高、中、低剂量组,模型组,通窍鼻炎片组,每组各10只。在造模成功后分别按相应剂量灌胃各组动物14d,薯蓣丸低、中、高剂量组分别为1.89g/kg、3.77g/kg、7.54g/kg,通窍鼻炎片组为0.56g/kg,空白组、模型组为等量0.9%氯化钠注射液。流式细胞法检测各组Foxp3水平。结果:Foxp3百分比空白组为(2.00±0.09)%,模型组为(1.06±0.10)%,通窍鼻炎片组为(1.53±0.20)%,薯蓣丸高剂量组为(1.65±0.07)%,薯蓣丸中剂量组为(1.43±0.07)%,薯蓣丸低剂量组为(1.12±0.06)%。其他5组与空白组比较,空白组、通窍鼻炎片组、薯蓣丸高及中剂量组与模型组比较,薯蓣丸高、低剂量组与通窍鼻炎片组比较,薯蓣丸中、低剂量组与高剂量组比较,薯蓣丸中剂量组与低剂量组比较,差异均有统计学意义(P<0.05);但薯蓣丸低剂量组与模型组比较,差异无统计学意义(P>0.05)。结论:薯蓣丸高、中剂量对豚鼠变应性鼻炎动物模型的Foxp3具有调节作用而低剂量作用不显著。

Objective ：To investigate the effect of Dioscorea opposita Thunb. pills on Foxp3 in guinea pigs with al- lergic rhinitis. Methods ： A total of 60 guinea pigs were randomly divided into blank group, high -, medium -, and low -dose Dioscorea opposita Thunb. pill groups ,model group,and Tongqiao Biyan tablet group,with 10 guinea pigs in each group. After the model was established successfully, the guinea pigs in the high - , medium - , and low -dose Dioscorea opposita Thunb. pill groups were given Dioscorea opposita Thunb. pills by gavage at doses of 1.89g/kg, 3.77g/kg, and 7.54g/kg, respectively,those in the Tongqiao Biyan tablet group were given Tongqiao Biyan tablets by gavage at a dose of 0. 56 g/kg, and those in the blank group and the model group were given 0. 9% sodium chloride injection of the same volume. Flow eytometry was performed to measure the level of Foxp3. Results ：The percentage of Foxp3 was（2. 00% ±0. 09） % in the blank group, （ 1.06% ±0. 10） % in the model group, （ 1.53%±0. 20） % in the Tongqiao Biyan tablet group, （1.65% ±0.07 ）% in the high -dose Dioscorea opposita Thunb. pill group, （1.43 % ± 0.07 ）% in the medium -dose Dioscorea opposita Thunb. pill group, and （1.12% ± 0.06）% in the low -dose Dioscorea opposita Thunb. pill group. There were significant differences between the blank group and the other five groups, between the model group and the blank group, the Tongqiao Biyan tablet group, the high - dose Dioscorea opposita Thunb. pill group, or the medium -dose Dioscorea opposita Thunb. pill group, between the high - and low - dose Dioscorea opposita Thunb. pill groups and the Tongqiao Biyan tablet group, between the medi- um- and low -dose Dioscorea opposita Thunb. pill groups and the high -dose Dioseorea opposita Thunb. pill group, and between the medium - and low - dose Dioscorea opposita Thunb. pill groups （P 〈 0. 05 ） ; there was no sig- nificant difference between the low - dose Dioscorea opposita Thunb. pill group and the model group （P 〉 0. 05 ）. Conelusion：High - and medium -dose Dioscorea opposita Thunb. pills can regulate Foxp3 in guinea pigs with allergic rhinitis, while low- dose Dioscorea opposita Thunb. pills do not have a marked effect.
