摘要 目的:探讨肥胖对小鼠各组织中Metrnl表达的影响。方法:高脂饮食4个月诱导小鼠肥胖,实时定量PCR与免疫组织化学法检测正常饮食与高脂饮食小鼠肝、骨骼肌、肾、结肠及脂肪组织中Metrnl mRNA与蛋白的表达。结果:高脂饮食增加了小鼠的体质量和白色脂肪量。同时,高脂饮食显著增加了白色脂肪组织Metrnl mRNA与蛋白的水平,但未增加肝、骨骼肌、肾、结肠组织中Metrnl的表达。结论:肥胖特异性增加脂肪组织中Metrnl的表达,从而形成"肥胖-脂肪组织上调Metrnl表达-代谢改善"的反馈调节通路。 Objective: To investigate the effect of obesity on the expression of Metrnl among different tissues. Methods: Mice were fed with high fat diet for four months to induce obese animal model. Metrnl mRNA and protein levels were detected wkh real time PCR and immunoh{stochemistry in the liver, skeletal muscle, kidney, colon and adipose tissue of mice fed with normal chow diet and high fat diet. Results: Both body weight and white adipose tissue weight were increased in high fat diet group. And Metrnl mRNA and protein levels in mice fed with high fat diet were dramatically increased in the white adipose tissue, but not in the liver, skeletal muscle, kidney and colon. Conclusion: Obesity can increase the expression of Metrnl in adipose tissue, thus forming the feedback pathway of ohesity-Metrnl upregulation in adipose tissue-metabolism improvement.
