蒙古族2型糖尿病合并脑卒中患者相关危险因素及其基因多态性的研究

Research of related risk factors and gene polymorphisms in Mongolian patients with type 2 diabetes mellitus complicated with stroke

目的探讨T2DM相关基因多态性与蒙古族糖尿病合并脑卒中患者的关系及相关危险因素分析。方法采用问卷调查和候选基因位点测序方法,将以往文献报道的34个T2DM易感基因位点作为目标进行测序,用SPSS 17.0软件分析临床特点以及相关的SNP位点基因型频率和等位基因频率。结果蒙古族T2DM合并脑卒中患者危险因素有年龄、胸围、LDL-C;相关SNP位点包括:TMEM163(rs6723108)(OR 8.6,95%CI 0.91~1.02,P=0.000)、PTPRD(rs17584499)(OR 2.96,95%C/1.08~4.85,P=0.000)及CENTD2(rs1552224)(OR 2.59,95%CI 1.43~4.70,P=0.020)。结论蒙古族T2DM患者发展为脑卒中,与患者年龄、胸围、LDL-C等危险因素密切相关;TMEM163(rs6723108)、PTPRD(rs17584499)和CENTD2(rs1552224)SNP位点的多态性与T2DM发生脑卒中密切相关。

Objective To investigate related risk factors and gene polymorphisms in Mongolian patients with type 2 diabetes mellitus （T2DM） complicated with stroke. Methods All the subjects in this study were surveyed by questionnaires. Candidate genes were tested by sequencing. A total of 34 candidate T2DM gene loci which were published previously were selected and sequenced. Clinical characteristics,genotypes and allele frequencies of all the SNPs were analyzed by SPSS 17.0. Results The risk factors for stroke in Mongolian patients with T2DM were age, bust, and LDL-C. The SNPs associated with stroke in patients with T2DM were as follows：TMEM163 （rs6723108） （OR 8.6,95%CI 0.91-1.02,P=0. 000）, PTPRD（rs17584499）（OR2.96,95%CI 1.08-4. 85,P=0. 000） and CENTD2 （rs1552224）（OR 2.59,95%CI 1.43-4.70,P=0. 020）. Conclusion Age,bust and LDL-C are the risk factors for stroke in Mongolian T2DM patients. The polymorphisms of TMEM163 （rs6723108）, PTPRD （rs17584499） and CENTD2（rs1552224） are closely related to T2DM complicated with stroke.
