摘要
目的探讨精氨酸加压素(AVP)对大鼠臂旁外侧核(LPB)使君子酸(AMPA)受体和N-甲基-D-门冬氨酸(NMDA)受体mRNA表达的影响。方法腹腔连续注射AVP1周后,采用RT-PCR技术,半定量分析大鼠LPB AMPA受体亚基GluR1、GluR2、GluR3和GluR4以及NMDA受体亚基NR2A和NR2BmRNA表达的变化。结果与对照组比较,AVP组大鼠LPB GluR1mRNA表达明显上调(P<0.05),而GluR2、GluR3和GluR4mRNA表达虽有上调趋势,但差异无统计学意义(P>0.05),NR2A和NR2BmRNA表达差异无统计学意义(P>0.05)。结论 AVP通过上调LPB AMPA受体,特别是GluR1亚基,而不是NMDA受体,增强LPB谷氨酸能突触传递。
Objective To investigate the effect of arginine vasopressin (AVP)on the subunit mRNA expressions of AMPA and NMDA receptors in lateral parabrachial nucleus (LPB)of rats.Methods The mRNA expressions of GluR1,GluR2,GluR3 and GluR4 in the AMPA receptors and those of NR2A and NR2B in the NMDA receptors were detected with RT-PCR after one-week intraperitoneal injection of AVP.Results Compared with the expressions in the control group,the mRNA expression of GluR1 in the experiment group was significantly upregulated by AVP in the LPB (P〈0.05),while the mRNA expressions of GluR2,GluR3,GluR4,NR2A and NR2B weren′t upregulated significantly (P〉0.05).Conclusion AVP enhances the glutamatergic transmission of LPB by upregulating AMPA receptors,especially GluR1 ,rather than NMDA receptors.
