miR-24基因家族进化及其在猪睾丸组织中的表达变化分析 被引量:4

摘要 通过miRBase数据库检索miR-24基因家族序列,利用Ensembl数据库信息确定miR-24家族在不同物种中的基因组定位,采用MEGA 5.0构建miR-24基因家族的系统进化树,并利用RT-qPCR检测miR-24-1及miR-24-2在沙子岭猪睾丸组织不同发育期(E90、D1、D30、D60、D90、D120、D150、D180)的表达变化。在miRBase数据库检索32个物种共66条序列,且各序列均具有高度保守性。基因组定位显示大部分物种的miR-24基因位于常染色体上。RT-qPCR结果显示,miR-24基因家族2个成员(miR-24-1、miR-24-2)在沙子岭猪睾丸不同发育阶段呈现出协同表达的规律。本研究对miR-24基因家族进行系统进化分析并检测其在沙子岭猪睾丸组织中表达变化规律,为进一步探明miR-24基因家族的功能及作用机制奠定基础。 The sequences of miR-24 gene family members in different animal species were searched from miRBase database, the miR-24 genes were located by Ensembl database, and its phylogenetic tree was constructed by MEGA 5.0, the expression levels of miR-24-1 and miR-24-2 were detec- ted at different developmental stages (E90, D1, D30, D60, D90, D120, D150, D180) of porcine testicular tissue by real-time fluorescence quantitative PCR. A total of 66 sequences in 32 species had been searched and all members of the miR-24 gene family were highly conserved. The analysis of localization showed that most of miR-24 genes were located on autosome. RT-qPCR results showed a co-expression of the miR-24-1 and miR-24-2 expression levels at different development stages. In this study, the evolutionary process of miR-24s and detected their expression patterns in the Shaziling pig testis was analyzed, which could provide the research basis for further study on the function and mechanism of miR-24 gene family.
