响应面优化秋茄叶粗多糖提取及其抗肿瘤活性 被引量:1

摘要 基于单因素对多糖提取率的影响,采用三因素三水平的响应面分析法优化秋茄叶粗多糖提取工艺,并采用MTT法测定其对H22肝癌细胞体外增殖抑制效果.秋茄叶粗多糖最佳提取条件为:超声温度68℃、时间90min、固液比1∶25,采用最佳提取工艺时秋茄叶粗多糖提取率为2.09%。秋茄叶粗多糖对H22肝癌细胞具有较强体外增殖抑制活性,且抑制活性与粗多糖浓度呈一定的正相关关系.该方法简单可行,粗多糖提取率较高且稳定,秋茄叶多糖对H22肝癌细胞具有体外增殖抑制效果. The extraction technology and inhibition of hepatoma carcinoma cell H22 in Vitro of crude polysaccharides from Kandelia candel were studied. Experimental factors and their levels were deter- mined by one-factor tests.Subsequently,the 3 factors and 3 levels' response surface method (RSM) test was designed to obtain the optimum extraction condition.The inhibitions of hepatoma carcinoma cell H22 in Vitro of crude polysaccharides were measured by MTT assay.The optimum extraction con- ditions to obtain the highest polysaccharides were 68℃, 90 min and solid-liquid ratio of 1 : 25,and the extracting rate of crude polysaccharides reached 2.09%.Polysaccharides from Kandelia candel could in- hibit cell proliferation of hepatoma carcinoma cell H22 in Vitro,and the inhibition rate increased with the polysaccharides concentration.The method is simple and practical with high yield of crude polysac- charities,and the polysaccharides from Kandelia candel can inhibit cell proliferation of hepatoma carci- noma cell H22 in Vitro.
