HPD酶在人肺腺癌细胞A549转移中的分子机制研究

摘要 目的分析酪氨酸代谢酶-羟苯丙酮酸二加氧酶(HPD)在肺腺癌细胞A549转移中的作用及其分子机制。方法在A549细胞株中高表达或下调HPD,分析其对肿瘤细胞转移的影响;通过transwell试验、划痕试验及荧光素酶试验等探索其相关分子机制。结果A549细胞中HPD表达水平升高能促进A549细胞转移,并升高肿瘤坏死因子-α(TNF-α)所引发的报告基因-核因-子κKB(NF-KB)的活性;HPD表达水平降低能抑制A549细胞转移,明显减少NF-κB的活性。在HPD高表达的细胞中基质金属蛋白酶-9(MMP-9)、连环蛋白(-catenin)mRNA水平显著增加,E-cadherin mRNA水平下降。结论HPD能促进肺腺癌细胞A549转移,该作用与细胞中HPD的表达水平密切相关,HPD能明显提高NF-κB的活性,并能调节多个转移相关基因。 Objective To analyze metabolic enzyme tyrosine hydroxyl benzene pyruvic acid oxygenase(hydroxyphenylpyruvate dioxy- genase, HPD) in lung adenocarcinoma A549 transfer of cancer cells and its molecular mechanism. Methods In A549 cell lines or cut HPD high expression, analyzes its effects on tumor cell metastasis. By transwell test, scratch test and luciferase test, etc. , to explore the related molecular mechanism. Results HPD expression in A549 cell levels can promote transfer of A549 cells, and elevated TNF alpha of report gene activity of NF-κB seem, HPD expression levels can inhibit A549 cell transfer,significantly reduce the re- port gene activity of NF-κB seem. In HPD high expression of cells, MMP-9 ,-catenin mRNA level increased significantly, E-cadherin mRNA levels drop. Conclusion HPD can promote gland cancer cells A549 lung metastasis,the effect is closely related to the cells of HPD expression level,HPD could significantly improve the activity of NF-KB seem report gene, and be able to adjust multiple metastasis associated genes.
