摘要
目的研究SLC39A6在宫颈癌中表达的临床意义及其对宫颈癌细胞功能的影响。方法基于GEO数据库中已有数据对SLC39A6在正常组织及肿瘤组织中的表达进行meta分析,继而在组织芯片上进行验证。通过TCGA数据库中数据对SLC39A6表达与宫颈癌患者的生存期进行相关性分析。通过RNAi技术结合慢病毒技术,对人宫颈癌细胞(Hela)中SLC39A6进行稳定干扰,研究SLC39A6基因干扰对宫颈癌细胞上皮间质转化(EMT)及转移能力的影响。结果通过GEO数据集和TCGA数据库中分析表明,宫颈癌组织中SLC39A6表达明显高于正常组织,且SLC39A6基因表达与患者总生存期及无病生存期呈负相关。干扰SLC39A6可明显抑制宫颈癌细胞上皮间质转化及转移能力。结论 SLC39A6与宫颈癌发生发展密切相关,SLC39A6可调控宫颈癌细胞转移,进而影响宫颈癌患者预后,表明SLC39A6可作为潜在的宫颈癌诊断标志物及治疗靶点。
Objective To investigate the expression of SLC39A6 and its effect on the cell function in cervical carcinoma.Methods The expression of SLC39A6 in cervical carcinoma was analyzed by the meta-analysis on the basis of the existing data in the GEO database and verified on the tissue chips.The TCGA database was used to analyze the relationship between the SLC39A6 expression and the survival time of patients with cervical cancer.RNAi combined with the lentivirus technology was used to investigate the effect of SLC39A6 on the epithelial mesenchymal transition(EMT)and the metastatic ability of cervical cancer cells by interfering with the stability of SLC39A6.Results The study of GEO and TCGA databases showed that the expression of SLC39A6 in the tumor tissues was significantly higher than that in the normal tissues,and it was negatively correlated with the survival time of patients with cervical cancer.The interference of SLC39A6 obviously inhibited the EMT and metastatic ability of cervical cancer cells.Conclusion SLC39A6 is closely related to the occurrence and development of cervical cancer.The inhibition of SLC39A6 can block the metastasis of cervical cancer cells and further affect the prognosis of patients with cervical cancer,which indicates that SLC39A6 can be used as a potential diagnostic marker for cervical cancer and the therapeutic targets.
