摘要
目的:建立同时测定通脉颗粒中丹参素、原儿茶酸、葛根素、大豆苷、阿魏酸、射干苷、丹酚酸B含量的方法。方法:采用高效液相色谱法测定。采用Agilent Zorbax SB-Aq色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸,梯度洗脱,流速1.2ml/min,柱温25℃。检测波长:丹参素、葛根素、大豆苷、丹酚酸B为287nm;原儿茶酸、射干苷为260nm;阿魏酸为322nm。结果:丹参素、原儿茶酸、葛根素、大豆苷、阿魏酸、射干苷、丹酚酸B的质量浓度分别在3.539~177.0μg/ml、0.3392~16.96μg/ml、2.755~137.8μg/ml、3.723~186.2μg/ml、1.699~84.96μg/ml、0.9440~47.20μg/ml、3.282~164.1μg/ml内与色谱峰峰面积呈良好的线性关系,平均回收率在92.4%~98.9%。结论:本方法简便、快速、可靠,可用于通脉颗粒的质量控制。
Objective: To establish method for the simultaneous determination of danshensu, protocatechuic acid, puerarin, daidzin, ferulic acid, belamcandin and Salvianolic acid B in Tongmai granules. Mthods : Samples were determined by HPLC on Agilent' s column ZORBAX SB - Aq (4.6 × 250mm,5 pan), which maintained at 25 ℃ column temperature, using 0. 1% (v/v) phosphoric acid - acetonitrile in a gradient elution mode as the mobile phase while the flow rate was 1.2ml/min. The detection wavelength were set at 287 nm for danshensu, puerarin, daidzin and Salvianolic acid B, 26Ohm for protocatechuic acid and belamcandin, 322nm for ferulic acid. Results: The linear ranges of danshensu, protocatecbuic acid, puerarin, daidzin, ferulic acid, belamcandin and Salvianolic acid B were 3. 539 - 177.0μg/ml,0. 3392 - 16. 96 μg/ml,2. 755 - 137. 8 μg/ml,3.723 - 186. 2 μg/ml, 1. 699 -84. 96 μg/ml,0. 9440 -47. 20 μg/ml,3. 282 - 164. 1 μg/ml, respectively. The average recoveries between 92. 4% - 98.9%. Conclusion: The method is simple, rapid and reliable, and it can be used for the quality control of Tongmai granules.
