摘要 目的:研究沙棘籽油中脂肪酸成分,定量分析并建立毛细管气相色谱法(CGC)测定沙棘籽油中维生素E(Vit E)含量的方法。方法:采用溶剂回流提取法提取沙棘油中挥发油成分,并用气相色谱-质谱联用法(GC-MS)对其进行定性、定量分析。采用岛津GC-2010 plus气相色谱仪,以正三十二烷为内标物对Vit E进行测定,并对该方法进行方法学考察。结果:从沙棘籽油中分离鉴定出28个化合物;采用Rtx-5毛细管色谱柱(30 m×0.25 mm,0.25μm);柱温为程序升温:150℃保持13 min,以10℃·min(-1)的升温速率升至300℃,保持8 min;FID检测器温度为300℃;进样口温度为310℃;无分流进样。在此条件下,Vit E和内标物分离良好,在0.02~0.43 mg·m L(-1)浓度范围内呈良好线性关系,r=0.999 6。结论:建立以CGC测定沙棘籽油中维生素E含量的方法,灵敏、准确、重复性好,可作为沙棘籽油中Vit E的含量测定方法。 Objective: To analyze the fatty acids composition in the oil from seed of Hippophae rhamnoides, and establish a capillary gas chromatography (CGC) method to determine the content of vitamin E in the seed oil of Hippophae rhamnoides. Methods: Oil was extracted from Hippophae rhamnoides seed by heated reflux solvent. GC-MS was adopted to determine its chemical constituents. The quantifcation of the vitamin E (Vit E) content was conducted by GC-2010 plus. Dotriacontane was acted as the internal standard. Results: Totally 28 chemical constituents were extracted. The GC system consisted of a Rtx-5 capillary column (30 m × 0.25 mm, 0.25 μm). Column temperature was temperature programming including 150 ℃ for 13 min, rose to 300 ℃ by the rate of 10 ℃·min-1 and maintained 300 ℃ for 8 min. The temperature of FID detector and the injection was 300 ℃ and 310 ℃ respectively. The splitless injection was adopted, Vitamin E was separated satisfactorily from dotriacontane. The calibration curve for Vitamin E was linear in the range of 0.02 – 0.43 mg·mL^-1 (r = 0.999 6). Conclusion: The capillary GC method was sensitive and reliable. It could be used to determine the content of vitamin E in the Seed oil of Hippophae rhamnoides.
