一株烟碱降解菌的筛选、保存及其降解性能 被引量:3

摘要 为了探究不同培养基保存方式、不同培养液条件下烟碱降解菌的降解性能,以烟碱为唯一碳源和氮源,从河南农业大学许昌校区现代烟草农业科教园区植烟土壤、腐烂烟叶中分离得到25株烟碱降解菌(耐受烟碱浓度为6 g/L),并选择降解效率较高的几株作为继续研究对象。研究表明:用烟碱作为唯一碳源和氮源的培养基(烟碱浓度2~3 g/L)斜面保存的菌株降烟碱活性较高;筛选得到一株烟碱高效降解菌5A-6-2,在其它碳、氮源同时存在时,仍然具有较强的烟碱降解能力;将5A-6-2接种至固体烟叶碎片中,28℃发酵7 d后,烟叶中烟碱含量下降了44.2%;通过16S r DNA序列分析发现,其与恶臭假单胞菌(Pseudomonas putida)同源性为99%。 In order to explore the different culture medium preservation methods, the degradation performance of nicotine-degrading bacteria under different culture conditions, with the nicotine regarded as the sole carbon source and nitrogen source, twenty-five nicotine-degrading bacteria are isolated from the tobacco-growing soil and rotten tobacco leaves of the modern tobacco agriculture science and education park of Xuchang Campus of Henan Agricultural University, and the nicotine concentration is 6 g/L, and several strains with high degradation efficiency are selected as the research object. The results show that the nicotine activity of the strain preserved with nicotine as the sole carbon source and nitrogen source (2~3 g / L nicotine) is higher. A nicotine-degrading bacterium 5A-6-2 is screened and still have strong ability of nicotine degradation when other carbon and nitrogen sources are present. 5A-6-2 is inoculated into solid flue debris, and the nicotine content of tobacco leaves decrease by 44.2% after 7 d fermentation at 28 ℃. 16S rDNA sequence analysis shows that it is 99% homology to Pseudomonas putida.
