摘要
为了解鄱阳湖水域的细菌群落多样性,选择鄱阳湖核心区具有代表性的15个点,运用16Sr DNA PCRDGGE分析技术结合化学分析方法进行分析,并将其部分结果与同种样品的高通量测序分析结果进行比较。结果表明:15个区域的水体均可分离出30到61种优势菌群,隶属于7门69属90种,各区域之间细菌群落相似性系数在49.0%~74.0%;PCR-DGGE与高通量测序分析方法相比较,PCR-DGGE方法只分析了样品中细菌群落数的1/10左右、看似与高通量方法中的丰度大于50的优势细菌群落数相接近,而且代表了样品总细菌群落丰度的91.7%,但2种方法的细菌多样性指数差异显著。主要环境指标(DO、电导率、ORP、NTU、PAR、矿化度和盐度)和营养盐(TN、TP、NO_2、NH_4、NO_3、PO_4)与细菌群落多样性指数之间存在一定的相关性,显示了鄱阳湖细菌群落对湖泊生境的适应性。
In order to explore the bacterial community diversity of Poyang Lake, in this study 15 typical sampling points were selected in the core area of Poyang Lake.16S rDNA PCR-DGGE combined with chemical measurement were applied to the analysis.Illumina sequencing approach was applied to the analysis of some samples and the results were compared with those from PCR-DGGE method.The results showed that a range of 30 to 61 dominant bacterial groups could be isolated from the water in 15 regions.These bacteria belong to 7 phyla,69 genera and 70 species.Similarity coefficient of bacterial communities among the regions was 49.0%- 74.0%.Comparing with the high-throughput sequencing method, the PCR-DGGE method only analyzed approx-imately ^^10 number of bacterial communities in the same samples.In PCR-DGGE method, the community was similar to the dominant bacterial groups with higher abundance of 50 which accounted for 91.7% abundance of the total community in high through sequencing method.Despite this similarity in analyzing dominant groups, the difference in bacterial diversity index between the two methods was significant. Furthermore, there were some correlations between the main environmental indicators ( DO, conductivity, ORP, NTU, PAR, salinity and salinity) and nutrients( TN, TP, NO2, NH4, NO3, PO4 ) and bacterial community diversity index, suggesting the adaption of bacterial communities to the environment in Poyang Lake.
