摘要
目的动态分析抗-HCV阳性献血者血浆miR-122的表达特点及与HCV感染诊断指标的关联。方法对133名抗-HCV阳性献血者、24名健康献血者及15名确证抗-HCV阳性追踪献血者血液标本抽提血浆miRNA,以实时荧光定量PCR法检测血浆miR-122,分析血浆miR-122相对表达量与HCV RNA、抗-HCV临界值指数(COI)等指标关联性。结果 ROC曲线分析:miR-122在抗-HCV+/HCV RNA+组和抗-HCV+/HCV RNA-组中诊断丙型肝炎的AUC(ROC曲线下面积)为0.830(95%CI:0.755-0.889),区分抗-HCV+/HCV RNA+组和抗-HCV+/HCV RNA-组的敏感性和特异性分别为74.7%和82.4%。追踪15例抗-HCV+/HCV RNA+献血者血浆miR-122表达明显降低(P<0.05)。结论血浆miR-122与HCV感染密切相关,确证HCV RNA感染献血者血浆miR-122随时间迁移表达降低。
Objective To dynamically monitor the expression of plasma miR- 122 in anti-HCV positive blood donors and analyze the relationship between miR-122 levels and HCV diagnostic test results. Methods 133 anti-HCV reactive donors, 24 healthy blood donors, 15 anti-HCV reactive and HCV RNA positive followed up blood donors were included in the study. The level of miR-122 in plasma were measured using real-time quantitative RT-PCR, and the relationship between miR-122 levels and HCV RNA, COI were statistically analyzed. Results From discriminating anti-HCV+/HCV RNA+ positive blood donors from those anti-HCV +/HCV RNA- negative people, the AUC-ROC ( the area under the ROC curve) was 0. 830 (95%CI :0. 755-0. 889) , and the sensitivity and specificity were 74. 7% and 82.4%, respectively. The expression of miR- 122 in plasma was significantly decreased in 15 anti-HCV+/HCV RNA+ patients (P〈 0. 05 ). Conclusion MiR-122 is closely related to HCV infection, and decreased with time in blood donors of HCV RNA infection.
