Effect of PLGA electrospun nanofiber scaffold on the osteogenic differentiation of rat bone marrow derived mesenchymal stem cells

摘要 目的:构建一种能促进骨髓间充质干细胞(BMSCs)成骨分化的组织工程支架材料,以期用于骨损伤的再生和修复。方法:采用静电纺丝技术制备聚乳酸-羟基乙酸共聚物(PLGA)电纺丝。大鼠骨髓间充质干细胞(BMSCs)进行活细胞染色后在电纺丝表面培养,观察细胞形态及增殖情况。加入成骨诱导剂,观察PLGA电纺丝表面培养对BMSCs成骨分化的影响。结果:BMSCs在培养皿表面上形态更为铺展,在PLGA电纺丝表面细胞形态更为细长。BMSCs在PLGA纳米电纺丝上的生长状况良好,与培养皿二维生长相比,在PLGA纳米电纺丝表面细胞增殖较慢。PLGA电纺丝表面可明显促进BMSCs向成骨细胞分化,ALP、OPN阳性细胞比率均明显高于培养皿二维生长细胞。结论:PLGA电纺丝可以促进BMSCs的成骨分化,在骨组织工程构建中具有重要价值。 Objective: To construct a tissue engineering stem cells (BMSCs) in order to regenerate and repair scaffold for osteogenic differentiation of bone mesechymal bone damage. Methods: Poly lactic-co-glycolic acid (PLGA) electrospun nanofiber scaffold was prepared by electrospinning technique. Rat BMSCs were cultured on the surface of the PLGA nanofibers, and cell proliferation and morphology were observed. Effect of PLGA on the osteogenic differentiation of BMSCs on the surface of PLGA nanofibers was observed by the addition of osteogenic inducer. Results:BMSCs cultured in culture dish exhibited favourable spreadability, but cells on PLGA nanofibers exhibited more slender morphology. The proliferation activity of BMSCs ceils on PLGA nanofibers was good. Compared with cultivation on 2D dish surface, the proliferation of BMSCs on the surface of PLGA nanofibers was slower. And, PLGA nanofibers surface could significantly promote the differentiation of BMSCs to osteoblasts. The positive cell ratio of ALP, OPN was more than those cultured on 2D surface. Conclusion: The PLGA nanofibers could promote the osteogenic differentiation of BMSCs, which has important value in bone tissue engineering.
