十全大补合剂联合顺铂对结肠癌荷瘤鼠肿瘤生长的调节作用 被引量:2

摘要 目的:研究十全大补合剂联合顺铂对结肠癌荷瘤鼠肿瘤生长的调节作用。方法:选择SPF级C57BL/6雄性小鼠作为实验动物,通过皮下注射结肠癌细胞的方式建立荷瘤模型小鼠并随机分为模型组(生理盐水干预)、顺铂干预组(10mg/kg顺铂干预)和联合干预组(10 mg/kg顺铂+3.9 mL十全大补合剂干预)。干预后5、10、15d时,测量肿瘤组织质量以及增殖相关分子、侵袭相关分子的表达量。结果:干预后5、10、15d时,顺铂干预组和联合干预组小鼠的肿瘤质量均显著低于模型组,联合干预组小鼠的肿瘤质量显著低于顺铂组;顺铂干预组和联合干预组肿瘤组织中TPX2、Rac1、cyclinD1、SLP-2、CHD1L、LSD1、N-cadherin的表达量显著低于模型组,RNF181、E-cadherin的表达量显著高于模型组;联合干预组肿瘤组织中TPX2、Rac1、cyclinD1、SLP-2、CHD1L、LSD1、N-cadherin的表达量显著低于顺铂干预组,RNF181、E-cadherin的表达量显著高于顺铂干预组。结论:十全大补合剂联合顺铂能够较顺铂单药更为有效的抑制结肠癌荷瘤鼠的肿瘤生长。 Objective:To study the regulating effect of Shiquandabu mixture combined with cisplatin on the tumor growth in colon cancer-bearing mice.Methods:SPF C57BL/6males mice were selected as the experimental animals,made into tumor-bearing models through subcutaneous injection of colon cancer cells and then randomly divided into model group(normal saline intervention),cisplatin intervention group(10mg/kg cisplatin intervention)and combined intervention group(10 mg/kg cisplatin + 3.9mL Shiquandabu mixture intervention).Five days,10 dand 15dafter intervention,the tumor tissue mass as well as the expression of proliferation-related molecules and invasion-related molecules was determined.Results:Five days,10 dand 15dafter intervention,the tumor mass of cisplatin intervention group and combined intervention group were significantly lower than that of model group,and the tumor mass of combined intervention group was significantly lower than that of cisplatin group;TPX2,Rac1,cyclinD1,SLP-2,CHD1 L,LSD1and N-cadherin expression in tumor tissue of cisplatin intervention group and combined intervention group were significantly lower than those of model group while RNF181 and E-cadherin expression were significantly higher than those of model group;TPX2,Rac1,cyclinD1,SLP-2,CHD1 L,LSD1and N-cadherin expression in tumor tissue of combined intervention group were significantly lower than those of cisplatin intervention group while RNF181 and E-cadherin expression were significantly higher than those of cisplatin intervention group.Conclusions:Shiquandabu mixture combined with cisplatin can more effectively inhibit the tumor growth in colon cancer-bearing mice than cisplatin monotherapy.
