强脉冲光自动脉冲联合甲硝唑口服对轻、中度痤疮患者血清炎症介质的影响 被引量:10

摘要 目的:研究强脉冲光自动脉冲联合甲硝唑口服对轻、中度痤疮患者血清炎症介质的影响。方法:选择2013年5月~2015年12月期间在我院诊断为轻中度痤疮的84例患者并随机分为激光组和甲硝唑组,分别接受强脉冲光自动脉冲联合甲硝唑口服治疗以及甲硝唑单药口服治疗。治疗前和治疗后1、2周时,采集血清并采用荧光定量PCR试剂盒测定Toll样受体(TLR)分子、NLRP炎性小体的表达量,采用酶联免疫吸附试剂盒测定炎性因子的含量。结果:治疗后2周和4周时,两组患者血清中白介素(IL)-1β、IL-8、IL-12、IL-18的含量以及TLR2、TLR4、NLRP3、ASC、Caspase-1的mRNA表达量均显著低于治疗前且激光组患者血清中IL-1β、IL-8、IL-12、IL-18的含量以及TLR2、TLR4、NLRP3、ASC、Caspase-1的mRNA表达量均显著低于甲硝唑组(P<0.05)。结论:强脉冲光自动脉冲联合甲硝唑口服对轻、中度痤疮患者体内模式识别受体TLR2、TLR4、NLRP3所介导的炎症反应具有显著抑制作用。 Objective:To study the effect of intense pulsed light autopulse combined with oral metronidazole on serum inflammatory mediators in patients with mild and moderate acne.Methods:84patients who were diagnosed with mild-to-moderate acne in our hospital between May 2013 and December 2015 were selected and randomly divided into laser group and metronidazole group who received intense pulsed light autopulse combined with oral metronidazole therapy and oral metronidazole monotherapy respectively.Before treatment as well as 1week and 2weeks after treatment,the serum was collected,fluorescent quantitative PCR kits were used to determine the expression of TLR molecules and NLRP inflammasome,and enzymelinked immunosorbent assay kits were used to determine the levels of inflammatory cytokines.Results:2weeks and 4weeks after treatment,serum IL-1β,IL-8,IL-12 and IL-18 levels as well as TLR2,TLR4,NLRP3,ASC and Caspase-1 mRNA expression of both groups of patients were significantly lower than those before treatment,and serum IL-1β,IL-8,IL-12 and IL-18 levels as well as TLR2,TLR4,NLRP3,ASC and Caspase-1mRNA expression of laser group were significantly lower than those of metronidazole group.Conclusion:Intense pulsed light autopulse combined with oral metronidazole has significant inhibitory effect on the inflammatory response mediated by pattern recognition receptors TLR2,TLR4 and NLRP3in patients with mild and moderate acne.
