三黄益肾胶囊对糖尿病大鼠肾脏保护作用研究

Study on the renal protecting effect of Sanhuang Yishen Capsule in diabetic rats

目的探讨三黄益肾胶囊对糖尿病大鼠肾脏的保护作用及机制。方法选取健康Wistar大鼠60只,随机选取10只作为正常组,余50只采用链脲佐菌素(STZ)腹腔注射方法制作糖尿病大鼠模型。选取40只造模成功大鼠随机分为模型组、三黄益肾胶囊低剂量组、三黄益肾胶囊中剂量组、三黄益肾胶囊高剂量组,每组10只。三黄益肾胶囊低、中、高剂量组分别给予三黄益肾胶囊0.4 g/(kg·d)、0.8 g/(kg·d)、1.6 g/(kg·d)灌胃,正常组和模型组每日给予同等剂量生理盐水灌胃,均每日1次,连续8周。检测各组大鼠空腹血糖、肌酐、尿素氮、胆固醇、三酰甘油、高敏C反应蛋白(hs-CRP)、肿瘤坏死因子-α(TNF-α)水平,计算肾质量/体质量指数(肾质量/体质量×1 000),光镜下观察各组大鼠肾脏组织病理学表现。结果灌胃8周后,各组空腹血糖水平与灌胃前比较差异均无统计学意义(P均>0.05),模型组及各给药组间空腹血糖水平比较差异均无统计学意义(P均>0.05);模型组及三黄益肾胶囊各剂量组血清TG、TC、SCr、BUN、hsCRP、TNF-α水平和肾质量/体质量指数均明显高于正常组(P均<0.05),体质量均明显低于正常组(P均<0.05);三黄益肾胶囊中、高剂量组血清SCr、BUN、hs-CRP、TNF-α水平和肾质量/体质量指数均明显低于模型组及三黄益肾胶囊低剂量组(P均<0.05),三黄益肾胶囊中、高剂量组间比较差异均无统计学意义(P均>0.05);三黄益肾胶囊各剂量组TG、TC水平及体质量与模型组比较差异均无统计学意义(P均>0.05)。三黄益肾胶囊中、高剂量组肾组织病理学改变较模型组明显改善。结论三黄益肾胶囊可能通过降低CRP、TNF-α水平,抑制炎症反应来发挥对糖尿病大鼠肾脏的保护作用。

Objective It is to observe the protective effect and mechanism of Sanhuang Yishen Capsule on the kidney of diabetic rats. Methods 60 healthy Wistar rats were selected, in which 10 ones as normal group, the other 50 ones were used to establish diabetic model by intraperitoneal injection of STZ. In the successful models 40 rats were selected and randomly divided into model group, low, medium and high dose of Sanhuang Yishen Capsule groups, each group had 10 rats. The rats in low, medium and high dose groups were given Sanhuang Yishen Capsule 0.4, 0. 8, 1.6 g/（ kg · d） respectively by garage, the ones in normal group and model group were given the same volume of normal saline. All the rats were treated once per day and for 8 weeks. Fasting blood glucose （FPG） , creatinine, blood urea nitrogen （BUN） , cholesterol, triglyeeride lipid, hs- CRP, TNF alpha were determined, kidney weight/body weight index （kidney weight/weight × 1 000） was calculated, and renal histopathological changes were observed under light microscope. Results After treatment for 8 weeks,no significant difference in FPG was found in each group compared with that before gavage（P 〉 0.05） , and there was no significant difference in FPG among every group too （P 〉 0. 05 ）. The levels of serum TG, TC, SCr, BUN, hs-CRP, TNF-α and kidney weight/body weight ratio were significantly higher while body weight was lower in model group and every Sanhuang Yishen Capsule group than those in normal group（P 〈0.05） ; the levels of serum SCr, BUN, hs-CRP, TNF-α and kidney weight/body weight ratio in medium and high dose of Sanhuang Yishen Capsule groups were significantly lower than those in model group and low dose of Sanhuang Yishen Capsule group （ P 〈 0.05 ） , no significant difference in these indexes was found between medium and high dose groups; there was no significant difference in TG, TC and body weight among every Sanhuang Yishen Capsule group and model group （ P 〉 0.05 ）. The improvements of histopathology in medium and high dose groups were significantly better than that in model group （P 〈 0.05）. Conclusion Sanhuang Yishen Capsule plays a protective effect on the kidney of diabetic rats maybe by down-regulating the levels of CRP, TNF-α, and inhibiting inflammatory response.
