24味中药对金黄色葡萄球菌生物膜作用的研究 被引量:11

Study on Twenty-four Traditional Chinese Medicines Against Staphylococcus aureus Biofilm in Vitro

摘要 目的从24味中药中筛选金黄色葡萄球菌生物膜的抑制剂。方法以微孔板置片法建立金黄色葡萄球菌生物膜体外模型;倍比稀释法测定24味中药水提液对金黄色葡萄球菌的最小抑菌浓度(MIC);微孔板法结合结晶紫染色法检测不同浓度提取液对金黄色葡萄球菌生物膜的形成与清除作用。结果体外培养24h时形成成熟生物膜;五倍子MIC最低,为0.39 mg/ml,其次为苏木12.5 mg/ml,黄芩50 mg/ml,丹参100 mg/ml;黄芩、毛冬青、五倍子提取液在1/2 MIC时对生物膜形成具有明显抑制作用;五倍子、黄芩、蜂房、毛冬青、蒲公英在2MIC时对生物膜清除作用效果显著。结论五倍子和黄芩两味中药水提液对金黄色葡萄球菌生长具有明显抑制作用,且对其生物膜的形成与清除作用显著。 Objective To screen effective inhibitors on staphylococcus aureus biofilm from twenty -four traditional Chinese medi- cines (TCM). Methods Microtiter plate -based model system was used in this study, and the biofilm was determined by micros- copy and crystal violet staining. Minimal inhibitory concentration (MIC) was determined by using the method of double dilution. Results Biofilm was formed after twenty - four hours. The MIC of Gallnut water extraction was 0.39 mg/mL, followed by Sappan- wood 12.5 mg/mL and Radix Scutellariae 50 mg/mL. Radix Scutellariae, Pubescent Holly root and Gallnut water extractions had obvious inhibitory effect on the formation of biofilm in 1/2 MIC. Gallnut, Radix Scutellariae, Hives, Pubescent Holly root and Dandelion had obvious removal effect on biofilm in 2 MIC. Conclusion Because of the inhibitory and removal effect on staphylo- coccus aureus biofilm, the water extractions of Gallnut and Radix Scutellariae were selected to do further study.
