新城疫病毒分离株real-time RT-PCR检测与其遗传变异特性分析

摘要 对肉鸡养殖场的疑似新城疫病例提取病毒核酸,采用便携式real-time RT-PCR仪T-COR4在2 h内检测到5株新城疫病毒,分别编号为RS1、RS2、HY3、LZ4、PL5。对确诊的阳性样品进行病毒增殖,血凝/血凝抑制试验,MDT、EID50、ICPI指数测定,F基因的遗传变异分析,结果显示,RS1、RS2、HY3株为新城疫基因Ⅶd型强毒株,LZ4、HY5株为新城疫基因Ⅱ型弱毒株。 Viral nucleic acids of suspected Newcastle disease virus (NDV) infection were extracted in broiler farms. 5 Newcastle disease virus (NDV) strains were detected with the real-time RT-PCR method in the portable T-COR4 platform within 2 h, named with RS1, RS2, HY3, LZ4 and PL5 respectively. Then the confirmed positive samples were detected with virus proliferation, HA/HI test, MDT, EID50, ICPI indexes determination, and F gene genetic variation analysis, The test results showed that RS1, RS2 and HY3 isolates were genotype VKd velogenic NDV, and LZA and HY5 were gonotype Ⅱ lentogenic NDV.
