一株抗高粱蚜芽孢杆菌的分离与鉴定 被引量:4

摘要 为获得抗高粱蚜的芽孢杆菌,从被高粱蚜侵害的高粱根际土壤、叶片及死亡蚜虫中分离出7株芽孢杆菌.毒力试验表明,菌株RA1402对蚜虫的校正死亡率最高,可达57.3%.对该菌株进行形态特征、生理生化及16S r DNA序列分析,初步鉴定为特基拉芽孢杆菌(Bacillus tequilensis).其特征为:细胞呈杆状,链状或成对排列,芽孢位于中部或近中部,无孢囊;不能将硝酸盐还原为亚硝酸盐,可分解淀粉,能利用木糖、阿拉伯糖和甘露醇.其16S r DNA序列与特基拉芽孢杆菌的相似性达到99.93%,在Gen Bank上的登录号为KR819163. In order to obtain antagonistic Bacillus sp. ofMelanaphis sacchari, 7 Bacillus sp. strains were isolated from rhizosphere soil, leaf of sorghum and dead Melanaphis sacchari. Virulence test indicates that the correction death rate of RA1402 to Melanaphis sacchari is 57.3%. By morphological, biochemical and physiological characteristic, and 16S rDNA sequence analysis of the strain, RA1402 is identified as Bacillus tequilensis. The cell is rod and occur either in pairs or chains, spors form centrally or paracentrally in non-swollen sporangia. The strains could decompose starch, utilized arabinose, mannitol and xylose, Nitrate could not be reduced into nitrite in the cell. The sequence alignment of 16S rDNA showes the homology of RA1402 and Bacillus tequilensis is 99.93%, the accession number in the GenBank is KR819163.
