摘要
目的了解安徽地区2011-2013年引起手足口病(hand-foot-mouthdisease,HFMD)发病的病原构成情况和人肠道病毒71型(humanenterovirus71,HEV71)分离株的基因特征。方法利用“中国疾病预防控制信息系统”,对2011-2013年报告的安徽省HFMD病例病原学特征进行统计分析。对从HFMD病例咽拭子标本中分离的26株HEV71进行VP1基因全长扩增和序列测定,使用生物软件DNASTAR7.1比对分析同源性和应用Mega4.1构建系统发生树。结果安徽省2011-2013年HFMD病原构成主要为HEV71,柯萨奇病毒A组16型(coxsaekievirusA16,CVA16)和其他肠道病毒,阳性检出率分别为28.74%、10.77%和15.18%。HEV71VP1区基因序列同源性比对分析和系统发生树构建表明,26株HEV71属于C4a基因亚型,与2008年安徽阜阳流行株(EU703812)相比亲缘关系较近,VPl区核苷酸和氨基酸同源性分别为92.9%-98.8%和90.3%~99.7%。结论安徽省2011-2013年HFMD的主要病原体为HEV71,其次为其他肠道病毒和CVA16。26株HEV71的基因型属C4a亚型,与中国大陆同期主要流行毒株基因型相一致。
Objective To understand the constituent of pathogens causing hand-food-mouth disease (HFMD) and the characterization of the VP1 genes of human enterovirus 71 strains (HEV71) in Anhui province, 2011-2013. Methods Descriptive epidemiologic method was used for statistical analysis of the data on epidemiological characteristics and pathogen detection results of HFMD cases in Anhui province reported from the National Infectious Disease Surveillance System, 2011 to 2013. The completed VP1 genes of 26 HEV71 strains isolated from throat swabs of patients were amplified by using reverse transcription-PCR (RT-PCR). The positive PCR products were sequenced. Homology of the VP 1 genes were analyzed with DNASTAR 7.1 software, and Phylogenetic tree was constructed with Mega 4.1 software. Results The constituent of pathogens causing HFMD were HEVT1, CVA16 and other enteroviruses in Anhui province during 2011 to 2013, with the positive rates of 28.74%, 10.77% and 15.18%, respectively. Phylogenetic analysis of the VP1 gene sequences of HEV71 strains showed that 26 HEV71 strains belonged to the genotype of C4a. The nucleotide acid of 26 strains isolated were closed to the strain of Fuyang (EU703812) in 2008. As compared with Fuyang (EU703812), the homology of nucleotide and amino acid sequences were 92.9% ~ 98.8% and 90.3% - 99.7%, respectively. ConclusionsThe main pathogen of HFMD in Anhui province during 2011-2013 was HEV71, followed by other enteroviruses and CVA16. 26 isolated strains of HEV71 were C4a subtype, which was consistent with genotype of the main epidemic strains in China during the same period.
