摘要
运用荧光猝灭光谱、分子对接等手段研究了表没食子儿茶素没食子酸酯(EGCG)与牛乳β-乳球蛋白之间的相互作用及作用机理。首先采用荧光光谱法研究了EGCG与β-LG的结合反应,并测定了结合常数,然后再利用Autodock 4.0软件对EGCG与β-LG的分子对接进行了研究。结果表明EGCG与β-LG形成复合物从而猝灭β-LG的内源荧光,其荧光猝灭机理符合静态机制,在17℃、37℃下的结合常数分别为2.313×104L/mol、1.507×10~4L/mol。热力学数据表明,EGCG与β-LG存在疏水作用、氢键及静电作用,分子对接结果与荧光猝灭结果基本吻合。
In the present work, the interaction of EGCG and β-1actoglobulin was studied by fluorescence quenching spectra and molecular docking. The results showed that these interactions result in the endogenous fluorescence quenching of β-lactoglobulin,which belongs to a static quenching mech- anism. The calculated bind constants were 2. 313 × 10^4 L/mol and 1. 507× 10^4 L/mol at 17 ℃ and 37 ℃, respectively. The results revealed that quercetin tended to bind with β-1actoglobulin mainly by hydrogen bond and hydrophobic interaction, as well as electrostatic force. The result of molecular docking was identical essentially with the flurescence quenching's.
