摘要
目的研究斑蝥酸钠维生素B_6诱导MG-63细胞凋亡的作用机制。方法应用不同浓度的斑蝥酸钠维生素B_6作用于MG-63细胞,用MTT比色法检测药物对细胞的增殖抑制作用;反转录聚合酶链反应(RT-PCR)检测存活素信使核糖核酸的表达。结果斑蝥酸钠维生素B_6在抑制MG-63细胞增殖方面有显著作用,其作用具有时间和剂量依赖性,抑制率可高达(68.56±5.21)%。RT-PCR显示可下调survivin基因表达。结论斑蝥酸钠维生素B_6抑制MG-63细胞增殖效果明显,其机制可能与下调survivin基因的表达有关。
Objective To study the mechanism of Sodium Cantharidate and vitamin B6 inducing apoptosis of MG-63 cells. Methods The different concentrations of Sodium Cantharidinate and vitamin B6 were applied in MG-63 cells, the anti-proliferative effect of drug for cells was detected by MTT eolorimetry; reverse transcription polymerase chain reac- tion (RT-PCR) was used to detect the expression of survivin messenger RNA. Results Sodium Cantharidate and vitamin B6 could inhibit the proliferation of MG-63 cells in a time and dose-dependent manner, and the inhibition rate could be as high as (68.56±5.21)%. RT-PCR showed downregulation of survivin gene expression. Conclusion The inhibitory effect of Sodium Cantharidate and vitamin B6 on the proliferation of MG-63 cells is obvious, and the mechanism may be relat- ed to down regulation of the expression of survivin.
