摘要
鸡皮肤表现出黄色主要是由于表皮层类胡萝卜素沉积的结果,类胡萝卜素被降解,鸡皮肤呈现白色。在鸡皮肤中主要由BCO2基因编码的β-胡萝卜素加氧酶2(beta-carotene oxygenase 2,BCO2)发挥其降解类胡萝卜素的作用。为了探究BCO2基因与鸡皮肤颜色的相关性,本研究选取黄皮肤鸡种-广西黄鸡、胡须鸡及白皮肤鸡种-青脚麻鸡、丝羽乌骨鸡通过分子克隆测序的方法对该基因进行单核苷酸多态性(Single Nucleotide Polymorphisms,SNP)扫描,发现在黄皮肤鸡种中的5’非编码区存在C/G突变,并通过PCR测序在各个鸡种中进行了检测,发现检测的黄皮肤鸡种中均存在该突变,证明BCO2基因的SNP与黄皮肤性状有相关性。
Chicken show yellow skin because of the deposition of carotenoids in skin. When carotenoids in the epidermis are degraded, the skin of chicken show white. Some researches have found out that carotenoids are mainly degraded by BCO2(beta-carotene oxygenase 2 in skin, BCO2), BCO2 gene encode this enzyme is an important candidate gene for regulating the deposition of carotenoids. In order to study the relevant between BCO2 and the colour of chicken skin, we choose Guang Xi huang chicken and Bearded Chicken as yellow skin chicken, Partridge Shank Chicken and silky fowl as white skin chicken. we scanned SNP(Single Nucleotide Polymorphisms) in our chicken, we found a C/G mutation in 5'UTR(untranslated regions) in yellow skin chicken. We also detected the mutation between other chicken breeds, result shows that all of yellow skin chicken we detected also have this mutation, suggested that SNP in BCO2 is related to yellow skin trait.
