摘要
目的研究黄芪苷对丙型肝炎病毒(HCV)的抑制作用。方法 MTT法检测不同浓度黄芪苷对Huh7.5.1细胞的抑制作用;间接免疫荧光试验检测不同浓度的黄芪苷、利巴韦林和干扰素(IFN)的抗HCV效果;Western blot法检测不同浓度黄芪苷、利巴韦林、IFN作用的Huh7.5.1细胞中HCV NS5A蛋白的表达情况。结果不同浓度黄芪苷对Huh7.5.1细胞的抑制率不同,并呈剂量依赖性。在最佳细胞密度1×10~4个/孔、最适MOI为1的条件下进行试验,利巴韦林对细胞的毒性呈剂量依赖性;IFN无明显的细胞毒性,其抗病毒滴度为7×10^(1.77) PFU/ml;黄芪苷的细胞毒性较小,其抗病毒滴度为7×10^(3.67) PFU/ml。黄芪苷能降低NS5A蛋白的表达,且随浓度增加抑制作用越明显。结论黄芪苷具有抗HCV的能力,其作用机制是通过抑制NS5A来实现的。
Objective To investigate the inhibitory effect of Astragalin on hepatitis C virus(HCV). Methods The inhibitory effect of Astragalin at various concentrations on Huh7. 5. 1 cells was determined by MTT assay, while that of various concentrations of Astragalin, ribavirin and interferon(IFN) on HCV by IFA. The expressions of HCV NS5 A protein in Huh7. 5. 1 cells treated with various concentrations of Asragalin, ribavirin and IFN were determined by Western blot. Results Astragalin showed dose-dependent inhibitory effect on Huh7. 5. 1 cells. Ribavirin showed dose-dependent toxicity to the cells at optimal density of 1 × 10~4 cells per well and optimal MOI of 1. IFN showed no significant cytotoxicity, of which antiviral titer was 7 × 10^(1.77)PFU/ml. Astragalin showed low cytotoxicity, of which the antiviral titer was 7 × 10^(3.67)PFU/ml. Astraglain decreased the expression of NS5 A, of which the inhibitory effect was dose-dependent.Conclusion Astragalin showed inhibitory effect on HCV by a mechanism of inhibiting NS5A.
