摘要
本文研究了溶解抑制剂的溶剂种类与滴加体积对酪氨酸酶荧光淬灭的影响。发现滴加抑制剂会降低体系中酪氨酸酶的浓度,有些溶剂本身也会直接导致酶荧光的淬灭。当每次滴加1μL,滴加20次对酪氨酸酶的荧光没有影响;随着溶剂含量的增加,丙酮及乙酸对酪氨酸酶荧光有明显的淬灭作用,甲醇、石油醚、乙酸乙酯则没有明显的影响。因此,在研究抑制剂与酪氨酸酶作用机制时可用甲醇、石油醚、乙酸乙酯溶解水溶性较差的抑制剂。
In this paper, the effects of solvent type and volume on the fluorescence quenching of tyrosinase were studied. It was found that the addition of inhibitors could decrease the concentration of tyrosinase in the system, and some of the solvents could directly cause the quenching of enzyme fluorescence. When ti- tration PPB 20 times of 1μL each time, which without affect the fluorescence of tyrosinase, so that titra- tion method can be used in the research of tyrosinase and inhibitor. Titration 20 times of 1 μL each times of methanol, petroleum ether or ethyl acetate without obvious effect on tyrosinase fluorescence, therefore, it is possible to use methanol and petroleum ether and ethyl acetate to dissolve the inhibitor with less water- soluble in the method of titration.
