摘要
目的:分离筛选两面针内生真菌Y118菌株及其代谢产物抗HepG2肿瘤细胞活性部位。方法:采用MTT比色法对两面针内生真菌Y118菌株及发酵代谢产物进行抗HepG2肿瘤细胞活性测定。结果:菌株Y118发酵后的菌丝体甲醇提取物,丙酮提取物和乙酸乙酯提取物浓度为80ug/ml时均对HepG2细胞有抑制作用。菌株的发酵液对HepG2细胞有明显的抑制作用。当浓度达到5%时,对HepG2细胞的抑制率达到43.94%;当浓度为10%时,对HepG2细胞的抑制率达到74.34%。结论:菌株Y118的甲醇提取物,丙酮提取物、乙酸乙酯提取物和发酵液均对HepG2细胞有抑制作用,其中发酵液的抑制作用较为明显。
Objective:To isolate and sift the Y118 strains of Zanthxylumnitidum(Roxb.)DC endophytic fungi and active sites of their metabolites against HepG2 tumor cells.Methods:MTT colorimetric assay was used to determine the activity of the Y118 strains of endophytic fungi and active sites of their metabolites against HepG2 tumor cells.Results:The methanol extract, acetone extract and ethyl acetate extract concentrations of Y118 strains of 80ug · ml^-1 after fermentation were inhibited by HepG2 cells.The fermentation liquid of the strains has obvious inhibitory effect on the HepG2 cells.When the concentration reaches 5%,the inhibition rate of HepG2 cells reaches 43.94%;When the concentration is 10%,the inhibition rate of HepG2 cells reaches 74.34%.Conclusion:The methanol extract, acetone extract,ethyl acetate extract concentrations of Y118 strains and fermentation liquid after fermentation have inhibitory effect on the HepG2 cells,and the inhibition of fermentation liquid is obvious.
