摘要
目的:研究卵巢癌的经阴道彩色多普勒超声血流参数与血管新生、癌细胞增殖活力的相关性。方法:收集在苏州大学附属常熟市第一人民医院妇产科手术切除Ⅰ~Ⅱ期卵巢癌41例、Ⅲ~Ⅳ期卵巢癌27例、卵巢良性病变病灶35例作为临床研究的样本,行经阴道彩色多普勒超声测定血流参数,经酶联免疫吸附实验测定血管新生基因、细胞增殖基因、抑癌基因的蛋白表达量。结果:Ⅰ~Ⅱ期卵巢癌、Ⅲ~Ⅳ期卵巢癌的RI、PI显著低于卵巢良性病变,Ⅲ~Ⅳ期卵巢癌的RI、PI显著低于Ⅰ~Ⅱ期卵巢癌;Ⅰ~Ⅱ期卵巢癌、Ⅲ~Ⅳ期卵巢癌病灶内VEGF165、NRP-1、SphK1、SphK2、YAP、CTGF、Gli2、TNFAⅠP8、LSD1的蛋白表达量显著高于卵巢良性病变,PTEN、MFN2、ST7L的蛋白表达量显著低于卵巢良性病变;Ⅲ~Ⅳ期卵巢癌病灶内VEGF165、NRP-1、SphK1、SphK2、YAP、CTGF、Gli2、TNFAⅠP8、LSD1的蛋白表达量显著高于Ⅰ~Ⅱ期卵巢癌且与RI、PI呈负相关,PTEN、MFN2、ST7L的蛋白表达量显著低于Ⅰ~Ⅱ期卵巢癌且与RI、PI呈正相关。结论:卵巢癌的经阴道彩色多普勒超声血流参数RI、PI的降低与血管新生增多、癌细胞增殖活力增强密切相关。
Objective: To study the correlation of transvaginal color Doppler ultrasound blood flow parameters of ovari- an cancer with angiogenesis and cancer cell proliferation activity. Methods~ A total of 41 cases of stage I-II ovarian cancer, 27 cases of stage Ⅲ-Ⅳ ovarian cancer and 35 cases of benign ovarian lesions surgically removed in Obstetrics and Gynecology De- partment of Changshu No. 1 People's Hospital Affiliated to Suzhou University between June 2014 and October 2016 were col- lected as clinical research samples, transvaginal color Doppler uhrasound was conducted to determine blood flow parameters, and enzyme-linked immunosorbent assay experiment was used to determine the protein expression of angiogenesis genes, cell proliferation genes and tumor suppressor genes. Results,, RI and PI of stage I-II ovarian cancer and stage Ⅲ-Ⅳ ovarian cancer were significantly lower than those of benign ovarian lesions, and the RI and PI of stage Ⅲ-Ⅳ ovarian cancer were significantly lower than those of stage Ⅰ-Ⅱ ovarian cancer; VEGF165, NRP-1, SphK1, SphK2, YAP, CTGF, Gli2, TNFAIP8 and LSD1 protein expression in stage Ⅰ-Ⅱ ovarian cancer and stage Ⅲ-Ⅳ ovarian cancer lesions were significantly higher than those in be- nign ovarian lesions while PTEN, MFN2 and ST?L protein expression were significantly lower than those in benign ovarian le- sions; VEGF165, NRP-1, SphK1, SphK2, YAP, CTGF, Gli2, TNFAIP8 and LSD1 protein expression in III-IV ovarian cancer lesions were significantly higher than those in stage I-Ⅱ ovarian cancer lesions and negatively correlated with RI and PI while PTEN, MFN2 and ST7L protein expression were significantly lower than those in FII and ovarian cancer lesions and pos- itively correlated with RI and PI. Conclusions: The decrease of transvaginal color Doppler ultrasound blood flow parameters RI and PI of ovarian cancer is closely related to the increase of angiogenesis and the enhancement of cancer cell proliferation activi-
