温针灸对膝骨关节炎大鼠行为学及血清IGF-1浓度的影响

The Influence ofWarm Needling Method on the Concentration of IGF-1 and Ethology of the Rats with Knee Osteoarthritis

目的:观察温针灸对膝骨关节炎大鼠行为学及血清胰岛素样生长因子1(IGF-1)的影响。方法:将40只雄性大鼠随机分为正常组、模型组、药物组和温针灸组,每组10只。正常组不予处理,其他3组采用木瓜蛋白酶关节腔注射法造模,造模后模型组不做处理,温针灸组给予温针灸治疗,药物组给予美洛昔康溶液灌胃,治疗结束后观察各组大鼠Lequesne MG膝关节评估级别及血清IGF-1含量变化情况。结果:Lequesne MG膝关节评估在局部反应、步态反应、关节活动和关节肿胀方面温针灸组与模型组相比差异有统计学意义(P<0.05);温针灸组血清IGF-1浓度较模型组和药物组升高,差异有统计学意义(P<0.05)。结论:温针灸可改善大鼠行为学功能,能提高大鼠血清中IGF-1表达水平,起到抗炎止痛,修复损伤的作用,是防治膝骨关节炎的可能机制之一。

Objective： To observe the effects of warm needling method on IGF-1 and the ethology of rats with knee osteoarthritis （KOA）. Methods： Forty rats were randomized into the normal group, the model group, the medicine group and warm needling group, ten rats each group. The normal group were unhandled, other three groups accepted intraarticular injection of papain to prepare the models, the warm needling group were treated by warm needling, and the medicine group accepted intragastfic administration of meloxicam tablets except the model group after the models were established successfully, the changes of IGF-1 contents and Lequesne MG knee assess- ment level of the rats in different groups were observed by the end of the treatment. Results： Lequesne MG knee as- sessment suggested that the different had statistical meaning in the comparisons of local reaction, gait response, joint mobility and joint swelling between warm needling group and the model group （P〈0.05）; the concentrations of serum IGF-1 raised when warm needling group was compared with the model group and the medicine group, and the difference showed statistical meaning （P〈0.05）. Conclusion： Warm needling could fight against the inflammation, stop the pain and repair the damage by improving rat ethology and raising IGF-1 expressions in rat serum, it is one of the potential mechanisms of preventing and treating KOA.
