摘要
目的探讨芹菜素对Wnt/β-catenin信号通路的调控及其对卵巢癌细胞侵袭能力的影响。方法在经芹菜素处理的人卵巢癌细胞系HO8910中,通过实时PCR和Western blot检测Wnt/β-catenin通路相关蛋白及下游蛋白的表达;采用Transwell法检测细胞体外迁移和侵袭能力;利用Luciferase双荧光素酶报告系统检测Wnt/β-catenin通路的活化情况。结果芹菜素作用24 h能够有效抑制HO8910细胞的迁移和侵袭能力,降低β-catenin和E-catenin的m RNA相对表达量和蛋白表达水平。芹菜素对Wnt/β-catenin信号通路中相关蛋白及下游靶蛋白具有调控作用(P<0.0001)。结论芹菜素可以通过抑制人卵巢癌细胞中Wnt/β-catenin信号通路,下调促转移靶蛋白的表达,抑制卵巢癌细胞的迁移和侵袭能力。
Objective To study the effects of apigenin on the invasion ability of ovarian cancer cells by the regulation of Wnt signaling pathway. Methods HO8910 cell line (Human ovarian cancer cells) was treated with apigenin. The Wnt/β-catenin signal pathway related proteins and downstream proteins were detected by real-time PCR and Western Blot. Transwell assay was used to detect the migration and invasion of cells in vitro, and the activation of Wnt/β-catenin pathway was detected by Luciferase double luciferase reporter system. Results Apigenin could inhibit the migration and invasion of HO8910 cells effectively after treatment for 24 h, and the relative mRNA expression and protein expression levels of β-eatenin and E-catenin reduced. Apigenin played roles in regulating related proteins and downstream tar- get proteins of Wnt/β-catenin signaling pathway (P〈 0.0001). Conclusion Apigenin can downregulate the expression of downstream target proteins of Wnt/β-catenin signaling pathway in human ovarian cancer cell line HO8910, and inhibit the migration and invasion abilities of ovarian cancer cells finally.
