不同砷暴露人群血清vitronectin表达的验证

Validation of serum vitronectin expression in different arsenic exposed population

摘要 目的验证蛋白质组学筛选出的慢性砷暴露血清差异表达蛋白——玻璃粘连蛋白(vitronectin)含量,为研究砷暴露人群血管疾病的发生提供线索。方法采用酶联免疫吸附试验(ELISA),验证筛选出的vitronectin在不同砷暴露剂量组的表达。结果对照组(228.45±34.14 ng/ml)和低(219.31±28.73 ng/ml)、高(199.84±29.11 ng/ml)剂量砷暴露组vitronectin血清含量差异有统计学意义(F=128.00,P<0.01),水砷含量与血清vitronectin水平呈负相关(OR=-383.95)。结论初步验证vitronectin可作为砷暴露致健康损害的血清生物标志物。 Objective To verify differentially expressed proteins of vitronectin related to vascular disease of human serum in chronic arsenic exposure, to provide clues for the study on vascular disease in arsenic exposed population by proteomics.Methods ELISA was taken to verify differentially expressed vitronectin in different arsenic exposure dose groups. Results There was statistical difference(F=128.00, P〈0.01) in serum levels of vitronectin in the control group(228.45±34.14ng/ml), the low(219.31±28.73 ng/ml) and high(199.84±29.11 ng/ml) dose arsenic exposure groups. The serum vitronectin content and the content of arsenic in water showed negative correlation(OR=-383.95). Conclusions Vitronectin is preliminarily verified as a biomarker of health damage induced by arsenic exposure.
